血红素氧合酶-1对视网膜光损伤的保护作用

【摘要】: 目的通过制备大鼠视网膜光损伤的动物模型,观察并分析血红素氧合酶-1(HO-1)对大鼠视网膜光损伤时的细胞保护作用。 方法分别以绿色荧光灯为光源制备光照组模型及光照前30min腹腔注射血晶素(Hemin)制备诱导组模型,以正常大鼠作为对照组,取大鼠视网膜组织,免疫组织化学染色方法观察HO-1在各组大鼠视网膜组织中的表达情况,以及通过流式细胞术检测各组大鼠视网膜细胞的凋亡率。 结果光照组大鼠在接受光照后较正常对照组大鼠视网膜HO-1的表达有所增加(p0.05),第一天即有表达,第五天时达高峰,随后逐渐下降,而其视网膜细胞的凋亡率表现出相同的变化趋势。光照前腹腔注射血晶素(Hemin)的诱导组大鼠视网膜HO-1的表达明显增高,且各时间点(1d、3d、5d、7d)的表达量均高于光照组(p0.05),而其视网膜细胞的凋亡率较光照组下降,同时间点(1d、3d、5d、7d)凋亡率均低于光照组(p0.05),而高于正常对照组(p0.05)。 结论血红素氧合酶-1对光损伤时的视网膜具有明显的细胞保护作用,且其表达量的水平受其诱导剂的影响而增加。
[Abstract]:Objective to observe and analyze the protective effect of heme oxygenase-1 (HO-1) on retinal light injury in rats. Methods the model of light group was made with green fluorescent lamp and the model of induced group was induced by intraperitoneal injection of hemagglutinin Heminin 30 minutes before illumination. The normal rats were used as the control group and the retinal tissues of the rats were taken out. Immunohistochemical staining was used to observe the expression of HO-1 in the retina of rats in each group and the apoptosis rate of retinal cells in each group was detected by flow cytometry. Results the expression of HO-1 in the retina of the illumination group was higher than that of the normal control group. The expression of HO-1 increased on the first day, reached its peak at 5th days, and then decreased gradually. The apoptosis rate of retinal cells showed the same change trend. The expression of HO-1 in retina of rats induced by intraperitoneal injection of hemagglutinin before illumination was significantly increased. The apoptotic rate of retinal cells was lower than that of light group, and the apoptotic rate of retinal cells was lower than that of light group at the same time. The apoptotic rate of retina cells was lower than that of light group (P 0.05), but higher than that of normal control group (P 0.05). Conclusion heme oxygenase-1 has a significant cytoprotective effect on retina after light injury, and its expression level is increased by its inducer.
