表皮生长因子受体抑制剂抑制大胶质细胞活化对急性高眼压大鼠视网膜神经节细胞丢失的影响

【摘要】:目的探讨表皮生长因子受体(epidermal growth factor receptor,EGFR)抑制剂抑制大胶质细胞活化对实验性急性高眼压大鼠视网膜神经节细胞(retinal ganglion cells,RGCs)丢失的影响。方法用健康成年SD大鼠18只,随机分为3组:空白对照组,不做任何处理;实验组采用前房灌注法制造急性高眼压模型,造模成功后,按6 mg·kg-1·d-1用量给予大鼠口服EGFR抑制剂AG1478;实验对照组造模成功后给予大鼠口服等量生理盐水。3组均于7 d处死大鼠,观察各组视网膜结构的变化及采用免疫组织化学法观察阳性节细胞的表达。结果通过免疫组化法发现,随着造模时间的延长,视网膜结构变得模糊不清,RGCs阳性染色细胞逐渐减少。空白对照组、实验组、实验对照组RGCs Thy-1阳性表达光密度值分别是:143.666 7±4.041 5、139.032 2±1.734 0和101.102 3±2.000 1。实验组和空白对照组相比,视网膜Thy-1阳性表达差异无统计学意义(P0.05),实验对照组和空白对照组比较,差异有统计学意义(P0.05),实验组与实验对照组视网膜Thy-1阳性表达,差异亦有统计学意义(P0.05)。结论 EGFR抑制剂通过抑制大胶质细胞的活化对实验性急性高眼压大鼠模型的RGCs有保护作用。
[Abstract]:Objective to investigate the effect of epidermal growth factor receptor (EGFR) inhibitor on the loss of retinal ganglion cells and retinal ganglion cells (RGCs) in experimental acute intraocular hypertension rats. Methods 18 adult Sprague-Dawley (SD) rats were used to investigate the effects of epidermal growth factor receptor inhibitor (EGFR) on the loss of retinal ganglion cells (RGCs) in rats with acute intraocular hypertension (AIOP). Three groups were randomly divided into three groups: the blank control group without any treatment, the experimental group using the anterior chamber perfusion method to make the acute intraocular pressure model, after the model was successfully made, Rats were given EGFR inhibitor AG1478 at the dosage of 6 mg 路kg-1 路d-1, and rats in the control group were killed at 7 days after the rats were given the same amount of saline. The changes of retinal structure and the expression of positive ganglion cells were observed by immunohistochemical method. The optical density of RGCs Thy-1 positive expression in the blank control group, experimental group and experimental control group was 143.6667 卤4.041 5,139.032 2 卤1.734 0 and 101.102 3 卤2.000 1 respectively. There was no significant difference in the positive expression of Thy-1 between the experimental group and the control group (P 0.05). There was a significant difference between the experimental control group and the blank control group (P 0.05). The positive expression of Thy-1 in the retina of the experimental group and the experimental control group was positive. Conclusion EGFR inhibitor has protective effect on RGCs of experimental acute intraocular hypertension rat model by inhibiting the activation of macroglia cells.
