P38MAPK、CARP在间歇低氧大鼠心肌重塑中的作用

【摘要】:摘要: 目的: 通过构建间歇低氧大鼠模型,检测模型大鼠心肌肥厚指数;观察大鼠心肌细胞形态学变化;测定心肌组织中p38MAPK、CARP蛋白表达量的变化,探讨间歇低氧对心肌重塑的影响。 方法: 选取健康雄性SD大鼠24只,随机分3组:间歇低氧4周组(CIH4)、间歇低氧8周组(CIH8),常氧组(NC)每组8只;将间歇低氧组大鼠放入低氧舱中,交替通入氮气与氧气,分别于4周、8周时处死CIH4组和CIH8组大鼠,用分析天平测定各组体重、左心室重量及左室肥厚指数;对大鼠心肌组织行HE染色,观察各组心肌组织形态学变化;用免疫组织化学法测定各组大鼠左心室p38MAPK、CARP蛋白表达,用灰度值表示其表达量。 结果 1.与NC组大鼠相比,CIH4、CIH8组大鼠心肌肥厚指数增加,CIH8组更为明显(P<0.05,,有统计学意义); 2.对大鼠左心室心肌进行形态学研究,显示CIH4、CIH8组心肌细胞排列紊乱、有部分心肌纤维化,CIH8组更为明显。 3.CIH4、CIH8组p38MAPK蛋白表达较NC组增加,CIH8组较CIH4组更为明显(P<0.05,有统计学意义); 4.CIH4、CIH8组CARP蛋白表达较NC组增加,CIH8组较CIH4组更为明显(P<0.05,有统计学意义); 5. p38MAPK与CARP两个指标采用Person相关分析,发现两者具有明显相关性。 结论: 1.间歇低氧可能为引起大鼠心肌重塑的诱因之一; 2. p38MAPK与CARP的表达在CIH4、CIH8组大鼠心肌组织中上调,且随着低氧暴露时间的延长表达量增加明显; 3. p38MAPK与CARP的表达具有相关性,提示两者的共同作用可能是间歇低氧大鼠发生心肌重塑的机制之一。
[Abstract]:Abstract:. Objective:. The myocardial hypertrophy index was measured, the morphological changes of myocardial cells were observed, and the expression of p38MAPKnCARP protein in myocardial tissue was measured to investigate the effect of intermittent hypoxia on myocardial remodeling. Methods:. Twenty-four healthy male SD rats were randomly divided into three groups: intermittent hypoxia group (4 weeks), intermittent hypoxia group (8 weeks), intermittent hypoxia group (8 rats), normoxic group (n = 8), and intermittent hypoxia group rats were placed in hypoxic chamber and alternately injected with nitrogen and oxygen. The rats of CIH4 group and CIH8 group were killed at 4 weeks and 8 weeks, the weight, left ventricular weight and left ventricular hypertrophy index of each group were measured by analytical balance, the myocardial tissue of rats was stained with HE, and the morphologic changes of myocardium were observed in each group. Immunohistochemical method was used to detect the expression of p38 MAPK- CARP protein in left ventricle of rats in each group, and the expression of p38 MAPK- CARP protein was expressed by gray value. Results. 1.Compared with the NC group, the myocardial hypertrophy index of the CIH4 + CIH8 group was significantly higher than that of the CIH8 group (P < 0.05). 2. The morphologic study of left ventricular myocardium in rats showed that the arrangement of myocardial cells in CIH4 + CIH8 group was disordered, and some myocardial fibrosis was more obvious in CIH8 group. 3. The expression of p38 MAPK protein in CIH4 + CIH8 group was significantly higher than that in CIH4 group (P < 0.05). 4. The expression of CARP protein in CIH4 + CIH8 group was significantly higher than that in CIH4 group (P < 0.05). 5. The correlation between p38 MAPK and CARP was analyzed by Person, and it was found that there was a significant correlation between p38 MAPK and CARP. Conclusion:. 1. Intermittent hypoxia may be one of the inducements of myocardial remodeling in rats. 2. The expression of p38 MAPK and CARP was up-regulated in the myocardial tissue of CIH4 + CIH8 group, and the expression of p38 MAPK and CARP increased significantly with the prolongation of hypoxia exposure time. 3. The expression of p38 MAPK is correlated with the expression of CARP, which suggests that the co-action of p38 MAPK and p38 MAPK may be one of the mechanisms of myocardial remodeling in intermittent hypoxic rats.
