UHRF1在喉鳞状细胞癌组织中的表达及临床意义

【摘要】:目的:UHRF1是一类与细胞增殖密切相关的核蛋白,在人类多种肿瘤的发生发展过程中扮演着重要作用。然而,UHRF1与喉鳞癌的关系未见报道,因此,我们研究UHRF1在喉鳞癌组织中的表达情况及其临床意义,为喉癌的发生发展及寻求新的治疗靶点提供实验依据。 方法: 1运用qRT-PCR分别检测60例喉癌组织和60例对应的癌旁正常组织中UHRF1mRNA表达水平; 2运用免疫组织化学法和western blot方法检测UHRF1蛋白在60例喉癌及60例对应的癌旁正常组织中的表达情况,分析其与喉癌临床病理参数之间的关系。 结果: 1qRT-PCR结果显示UHRF1mRNA在喉鳞癌组织中的表达水平明显高于癌旁正常组织,表达差异具有显著统计学意义(P0.001); 2免疫组织化学及western blot结果显示UHRF1蛋白在78.3%(47/60)的喉癌组织高表达,而癌旁正常组织未见UHRF1蛋白表达,表达差异具有显著统计学意义(P0.001); 3UHRF1蛋白高表达与患者吸烟(P0.001)、较晚的T分期(P=0.005)及临床分期(P=0.044)、较差的组织学分化(P=0.048)具有相关性,而与患者年龄、性别及有无颈部淋巴结转移无关(P0.05)。 结论:UHRF1在喉鳞癌组织中表达上调,,可能与喉癌的发生发展有着密切关系,其可能成为判断喉癌生物学行为的重要指标。
[Abstract]:Objective: UHRF1 is a kind of nuclear protein closely related to cell proliferation, which plays an important role in the occurrence and development of various human tumors. However, the relationship between UHRF1 and laryngeal squamous cell carcinoma has not been reported. We studied the expression of UHRF1 in laryngeal squamous cell carcinoma (LSCC) and its clinical significance in order to provide experimental evidence for the occurrence and development of laryngeal carcinoma and seek for new therapeutic targets. Methods:. 1the expression of UHRF1mRNA in 60 cases of laryngeal carcinoma and 60 cases of adjacent normal tissues were detected by qRT-PCR. 2Immunohistochemical method and western blot method were used to detect the expression of UHRF1 protein in 60 cases of laryngeal carcinoma and 60 cases of adjacent normal tissues, and to analyze the relationship between the expression of UHRF1 protein and the clinicopathological parameters of laryngeal carcinoma. Results:. 1qRT-PCR results showed that the expression of UHRF1mRNA in laryngeal squamous cell carcinoma tissues was significantly higher than that in adjacent normal tissues, and the difference was statistically significant (P 0.001). 2the results of immunohistochemistry and western blot showed that the expression of UHRF1 protein was high in the laryngeal cancer tissues of 78.3% and 47 / 60%, but no expression of UHRF1 protein was found in the adjacent normal tissues. The difference was statistically significant (P 0.001). The high expression of 3UHRF1 protein was correlated with smoking, late T stage (P 0.005) and clinical stage (P < 0.044), and poor histological differentiation (P < 0.048), but not with age, sex and cervical lymph node metastasis (P 0.05). Conclusion the upregulation of the expression of WUHRF1 in laryngeal squamous cell carcinoma may be closely related to the occurrence and development of laryngeal carcinoma, and it may be an important index to judge the biological behavior of laryngeal carcinoma.
