应激反应对视网膜色素上皮细胞的影响

【摘要】:目的研究应激反应时大鼠血清中肾上腺素浓度的变化,并通过观察大鼠视网膜色素上皮细胞在应激状态下超微结构的改变,探讨应激反应与视网膜色素上皮细胞改变的相关性,为进一步研究视网膜疾病的发生发展奠定理论基础。 方法将24只健康雄性Wistar大鼠分成4组建立应激大鼠模型,分别予以束缚应激,诱导应激反应发生。收集大鼠血清样本后通过酶联免疫吸附法(ELISA),监测大鼠血清中肾上腺素浓度变化,整理数据,使用SPSS13.0统计学软件进行数据处理,并采用光镜、透射电镜技术对大鼠视网膜组织超微结构进行观察,从而得出结论。 结果 (1).对照组大鼠血清肾上腺素浓度在正常饲养阶段并未发生明显的变化,差异无统计学意义(P0.05)。 (2).适应组大鼠从实验第一周开始应激,与第一周初相比,血清肾上腺素浓度明显升高,差异有统计学意义(P0.05),第三周初停止应激,从第三周末血清肾上腺素浓度开始下降,但是较第一周初仍有升高。 (3).适应+应激组大鼠从实验第一周开始应激,与第一周初相比,血清肾上腺素浓度明显升高,差异有统计学意义(P0.05),并且随着应激时间的延长血清肾上腺素浓度逐渐升高。 (4).应激组大鼠从实验第三周开始应激,与第一周初相比,血清肾上腺素浓度明显升高,差异有统计学意义(P0.05),并且随着应激时间的延长血清肾上腺素浓度逐渐升高。 (5).普通光学显微镜观察:RPE细胞未见明显异常。透射电子显微镜观察(JEM-1200EX):RPE细胞核形状多不规则,细胞内多种细胞器发生病理性改变,线粒体空化肿大,可见色素颗粒,偶见内质网轻度扩张,且随着肾上腺素浓度增加而愈发严重。 结论 (1).持续应激可能使大鼠体内肾上腺素水平较应激前明显增高,并且随着应激时间的延长血清肾上腺素浓度逐渐升高。 (2).终止应激后大鼠体内肾上腺素水平下降但仍较应激前增高。 (3).应激状态下,肾上腺素浓度增高诱导RPE细胞内出现缺氧性改变,并随着应激时间的延长RPE细胞损害加重。
[Abstract]:Objective to study the changes of serum epinephrine concentration in rats during stress, and to investigate the relationship between stress response and retinal pigment epithelial cells by observing the ultrastructural changes of retinal pigment epithelial cells under stress. To lay a theoretical foundation for the further study of the occurrence and development of retinal diseases. Methods 24 healthy male Wistar rats were divided into 4 groups to establish stress model and were subjected to restraint stress respectively. After collecting rat serum samples, the changes of epinephrine concentration in serum of rats were monitored, the data were processed by SPSS13.0 software, and light microscope was used. The ultrastructure of rat retina was observed by transmission electron microscopy (TEM). Results. The serum epinephrine concentration in the control group did not change significantly in the normal feeding stage, but the difference was not statistically significant (P 0.05). The rats in the adaptation group began to stress from the first week of the experiment. Compared with the beginning of the first week, the concentration of serum epinephrine increased significantly, the difference was statistically significant (P 0.05). At the beginning of the third week, the stress was stopped, and the concentration of serum adrenaline began to decrease at the end of the third week. But it's still up from the beginning of the first week. Compared with the first week, the concentration of serum epinephrine in the adaptation stress group was significantly higher than that in the first week, the difference was statistically significant (P 0.05), and the concentration of serum epinephrine gradually increased with the prolongation of the stress time. The rats in the stress group began to stress from the third week of the experiment. Compared with the first week, the concentration of serum epinephrine increased significantly, the difference was statistically significant (P 0.05), and the concentration of serum epinephrine gradually increased with the prolongation of the stress time. Under ordinary optical microscope, there was no obvious abnormality in the RPE cells. Transmission electron microscopy (TEM) showed that the nuclei of JEM-1200EX1: RPE were irregular in shape, many organelles in the cells had pathological changes, mitochondria were cavitated and swollen, and pigment granules could be seen. Occasionally, endoplasmic reticulum dilated slightly, and became more serious with the increase of epinephrine concentration. Conclusion. The level of epinephrine in rats was obviously higher than that before stress, and the concentration of serum epinephrine increased gradually with the prolongation of stress time. After termination of stress, the level of adrenaline in rats decreased, but it was still higher than that before stress. Under stress, the increased concentration of epinephrine induced hypoxic changes in RPE cells, and the damage of RPE cells increased with the prolongation of stress time.
