BRMS1和SATB1在喉鳞状细胞癌中的表达及临床意义

【摘要】:目的:探讨乳腺癌转移抑制基因1(BRMS1)和特异AT序列结合蛋白1(SATB1)在喉鳞状细胞癌癌组织及癌旁正常组织中的表达情况、相关性及临床意义。 方法:1.采用免疫组化EliVision法检测20例声门上型喉癌和20例声门型喉癌癌组织及相应的癌旁正常组织中BRMS1和SATB1的表达情况。2.采用逆转录聚合酶链式反应(RT-PCR)检测20例声门上型喉癌和20例声门型喉癌癌组织及相应的癌旁正常组织中BRMS1mRNA和SATB1mRNA的表达情况。3.分析BRMS1和SATB1在喉鳞状细胞癌中表达的相关性及与患者的性别、年龄、临床TNM分期、分化程度和淋巴结转移的关系。 结果: 一.免疫组化结果 1.声门上型喉鳞状细胞癌癌组织中的BRMS1蛋白阳性表达低于癌旁正常组织的阳性表达,两组差异有统计学意义(P<0.05)。 2..声门型喉鳞状细胞癌癌组织中的BRMS1蛋白阳性表达低于癌旁正常组织的阳性表达,两组差异有统计学意义(P<0.05)。 3.声门上型喉癌组织中BRMS1的蛋白阳性表达与声门型喉癌组织中BRMS1的阳性表达的差异无统计学意义(P>0.05)。 4.声门上型喉鳞状细胞癌癌组织中的SATB1蛋白阳性表达高于癌旁正常组织的阳性表达,两组差异有统计学意义(P<0.05)。 5..声门型喉鳞状细胞癌癌组织中的SATB1蛋白阳性表达高于癌旁正常组织的阳性表达,两组差异有统计学意义(P<0.05)。 6.声门上型喉癌组织中SATB1蛋白的阳性表达与声门型喉癌组织中SATB1蛋白的阳性表达差异无统计学意义(P>0.05)。 7.声门上型喉癌中,BRMS1蛋白与SATB1蛋白的表达与TNM分期、肿瘤分化程度、有无淋巴结转移有关,与年龄、性别无关。 8.声门型喉癌中,BRMS1蛋白与SATB1蛋白的表达与TNM分期、肿瘤分化程度有关,与年龄、性别、有无淋巴结转移无关。 9.在声门上型喉癌和声门型喉癌中,BRMS1蛋白和SATB1蛋白表达均成负相关性。 二、RT-PCR结果 1.在声门上型和声门型组中,BRMS1mRNA在喉鳞状细胞癌中的表达均低于癌旁正常组织的BRMS1mRNA表达,两组差异有统计学意义(P<0.05)。 2.在声门上型和声门型组中,SATB1mRNA在喉鳞状细胞癌中的表达均高于癌旁正常组织的SATB1mRNA表达,两组差异有统计学意义(P<0.05)。 结论: 1.BRMS1在声门上型喉癌和声门型喉癌组织中的表达水平均低于癌旁正常组织。 2.SATB1在声门上型喉癌和声门型喉癌组织中的表达水平均高于癌旁正常组织。 3.声门上型喉癌中,BRMS1和SATB1的表达与TNM分期、肿瘤分化程度、淋巴结转移有关,与患者的年龄、性别无关。 4.声门型喉癌中,,BRMS1和SATB1的表达与TNM分期、肿瘤分化程度有关,与患者的年龄、性别、淋巴结转移无关。 5.BRMS1和SATB1的表达呈负相关,对BRMS1和SATB1进行联合检测可能成为判断喉鳞状细胞癌淋巴结转移情况的重要指标。
[Abstract]:Objective: to investigate the expression, correlation and clinical significance of metastasis suppressor gene 1 (BRMS1) and specific AT sequence binding protein (1) SATB1) in laryngeal squamous cell carcinoma (LSCC) and adjacent normal tissues. Methods the expression of BRMS1 and SATB1 in 20 cases of supraglottic laryngeal carcinoma and 20 cases of glottic carcinoma and their adjacent normal tissues were detected by immunohistochemical EliVision method. 2. Reverse transcriptase polymerase chain reaction (RT-PCR) was used to detect the expression of SATB1 in 20 cases of supraglottic laryngeal carcinoma and 20 cases of normal tissues adjacent to glottic carcinoma. Expression of BRMS1mRNA and SATB1mRNA in supraglottic laryngeal carcinoma, 20 cases of glottic laryngeal carcinoma and the corresponding adjacent normal tissues. 3. To analyze the correlation between BRMS1 and SATB1 expression in laryngeal squamous cell carcinoma and the sex of the patients. Age, clinical TNM stage, degree of differentiation and lymph node metastasis. Results:. I. Immunohistochemical results. 1. The positive expression of BRMS1 protein in supraglottic laryngeal squamous cell carcinoma was significantly lower than that in adjacent normal tissues (P < 0.05). 2.The positive expression of BRMS1 protein in glottic laryngeal squamous cell carcinoma was significantly lower than that in adjacent normal tissues (P < 0.05). 3. The positive expression of BRMS1 protein in supraglottic laryngeal carcinoma was not significantly different from that of BRMS1 in glottic laryngeal carcinoma (P > 0.05). 4. The positive expression of SATB1 protein in supraglottic laryngeal squamous cell carcinoma was significantly higher than that in adjacent normal tissues (P < 0.05). 5.The positive expression of SATB1 protein in glottic laryngeal squamous cell carcinoma was higher than that in adjacent normal tissue (P < 0.05). 6. The positive expression of SATB1 protein in supraglottic laryngeal carcinoma was not significantly different from that in glottic laryngeal carcinoma (P > 0.05). 7. The expression of BRMS1 protein and SATB1 protein in supraglottic laryngeal carcinoma was related to TNM stage, tumor differentiation, lymph node metastasis, age and sex. 8. The expression of BRMS1 protein and SATB1 protein in glottic laryngeal carcinoma was related to TNM stage, tumor differentiation, age, sex, lymph node metastasis or not. 9. The expression of BRMS1 protein and SATB1 protein in supraglottic laryngeal carcinoma and glottic laryngeal carcinoma were negatively correlated. RT-PCR results. 1. In supraglottic type and glottic type group, the expression of BRMS1mRNA in laryngeal squamous cell carcinoma was lower than that in adjacent normal tissue (P < 0.05). 2. The expression of SATB1mRNA in the supraglottic type and glottic type was higher in laryngeal squamous cell carcinoma than in the adjacent normal tissue (P < 0.05). Conclusion:. The expression of 1.BRMS1 in supraglottic laryngeal carcinoma and glottic laryngeal carcinoma was lower than that in adjacent normal tissues. The expression of 2.SATB1 in supraglottic laryngeal carcinoma and glottic laryngeal carcinoma was higher than that in adjacent normal tissue. 3. The expression of BRMS1 and SATB1 in supraglottic laryngeal carcinoma was related to TNM stage, tumor differentiation, lymph node metastasis, age and sex. 4. The expression of BRMS1 and SATB1 in glottic laryngeal carcinoma was related to TNM stage, tumor differentiation, age, sex and lymph node metastasis. The expression of 5.BRMS1 and SATB1 was negatively correlated, and the combined detection of BRMS1 and SATB1 might be an important index to judge the lymph node metastasis of laryngeal squamous cell carcinoma (LSCC).
