HIF-1α和c-met在喉鳞癌中的表达及其与微血管密度的关系

【摘要】:目的 观察缺氧诱导因子-1α (hypoxia-inducible factor1alpha, HIF-la)、肝细胞生长因子受体(hepatocyte growth factor receptor, c-met)和微血管密度(microvesel density, MVD)在人喉鳞状细胞癌的表达及联系,探讨HIF-1α和c-met在喉鳞状细胞癌发生发展和淋巴转移中的作用,从而为喉癌的发生机制与基因治疗提供一定的理论依据和实验基础。 方法 收集郑州大学第一附属医院2006~2010年耳鼻咽喉-头颈外科所做喉鳞癌标本58例,采用链霉菌抗生物素蛋白过氧化物酶(streptavidin peroxidase, SP)法免疫组织化学技术,检测HIF-1α和c-met在喉鳞状细胞癌不同分化类型中的表达,并测定微血管密度(MVD),观察HIF-1α、c-met和MVD之间的联系。 结果 HIF-1α阳性表现为棕褐色颗粒,主要定位在细胞核,部分在细胞质内。其在高、中、低分化喉癌中阳性区平均积分光密度(the average integrated optical density, AIOD)分别为121.91±3.59、133.31±2.65、146.76±6.61,经方差分析结果显示:不同分化类型间HIF-1α的差异有统计学意义(P0.05)。在颈淋巴结转移组与未转移组中阳性区AIOD分别为142.85±7.48、126.73±5.82,经t检验结果显示:两组的差异有统计学意义(P0.05);c-met阳性表现为棕黄色颗粒,主要定位在胞浆及胞膜。其在高、中、低分化喉癌中阳性区AIOD分别为120.23±3.04、131.04±4.60、159.12±9.26,经方差分析结果显示:不同分化类型间c-met的差异有统计学意义(P0.05)。在颈淋巴结转移组与未转移组中阳性区AIOD分别为149.88±14.85、124.81±5.87,经t检验结果显示:两组的差异有统计学意义(P0.05);高、中、低分化喉癌中MVD分别为11.82±2.38、23.08±4.95、39.44±4.30,经方差分析结果显示:不同分化类型间MVD的差异有统计学意义(P0.05)。在颈淋巴结转移组与未转移组中MVD分别为34.76±7.47、16.36±5.47,经t检验结果显示:两组的差异有统计学意义(P0.05)。Pearson相关性分析方法分析结果显示:HIF-1α与c-met之间的相关系数r=0.929,P0.001,有统计学意义,说明HIF-1α与c-met呈明显正相关;HIF-1α与MVD之间的相关系数r=0.967,P0.001,有统计学意义,说明HIF-1α与MVD也呈明显正相关;c-met与MVD之间的相关系数r=0.921,P0.001,有统计学意义,说明两者也呈正相关。 结论 HIF-1α、c-met和新生血管的形成在喉鳞状细胞癌的发生、发展、淋巴结转移中起重要作用,HIF-1α、c-met存在有协同作用。HIF-1α、c-met可能作为治疗喉鳞癌的潜在靶点。
[Abstract]:PurposeTo investigate the expression and association of hypoxia-inducible factor-1 alpha-hypoxia-inducible factor-1 (HIF-lalaa), hepatocyte growth factor receptor (c-metr) and microvesel density (MVD) in human laryngeal squamous cell carcinoma (LSCC), and to explore the role of HIF-1 伪 and c-met in laryngeal squamous cell carcinoma (LSCC) development and lymphatic metastasis.It provides a theoretical and experimental basis for the pathogenesis and gene therapy of laryngeal carcinoma.MethodFifty-eight cases of laryngeal squamous cell carcinoma (SCC) from 2006 to 2010 in the first affiliated Hospital of Zhengzhou University were collected. Streptavidin peroxidase (SP) immunohistochemical technique was used.The expression of HIF-1 伪 and c-met in different differentiation types of laryngeal squamous cell carcinoma (LSCC) was detected, and the microvessel density (MVD) was measured. The relationship between HIF-1 伪 c-met and MVD was observed.ResultThe expression of HIF-1 伪 was brown granules, mainly located in the nucleus and partly in the cytoplasm.The mean integral optical density of the average integrated optical was 121.91 卤3.59133.31 卤2.65146.76 卤6.61 in high, middle and low differentiated laryngeal carcinoma, respectively. The results of ANOVA showed that there were significant differences in HIF-1 伪 among different differentiation types.The AIOD of positive region in cervical lymph node metastasis group and non-metastasis group was 142.85 卤7.48126.73 卤5.82, respectively. The results of t test showed that the difference between the two groups was statistically significant (P 0.05). The positive rate of c-met was brown granules, mainly located in cytoplasm and cell membrane.The AIOD in high, middle and low differentiated laryngeal carcinoma was 120.23 卤3.04131.04 卤4.60159.12 卤9.26, respectively. The results of variance analysis showed that the difference of c-met between different differentiation types was statistically significant (P 0.05).The AIOD of positive region in cervical lymph node metastasis group and non-metastasis group were 149.88 卤14.85124.81 卤5.87, respectively. T test results showed that the difference between the two groups was statistically significant (P 0.05);The MVD in poorly differentiated laryngeal carcinoma was 11.82 卤2.38 卤23.08 卤4.95 卤39.44 卤4.30, respectively. The results of variance analysis showed that there was significant difference in MVD between different differentiation types.There was a significant positive correlation between HIF-1 伪 and c-met and the correlation coefficient between HIF-1 伪 and MVD, which was statistically significant. The correlation coefficient between HIF-1 伪 and MVD was also positively correlated with MVD (r = 0.921, P 0.001), which indicated that there was a positive correlation between HIF-1 伪 and MVD, which indicated that there was a positive correlation between HIF-1 伪 and MVD, which indicated that there was also a positive correlation between HIF-1 伪 and MVD.ConclusionHIF-1 伪 c-met and neovascularization play an important role in the development and development of laryngeal squamous cell carcinoma. HIF-1 伪 c-met may be a potential target for the treatment of laryngeal squamous cell carcinoma.
