鼻咽癌microRNAs差异表达谱的鉴定

【摘要】:目的:筛选及验证鼻咽癌中microRNAs(miRs)的差异表达谱。方法:通过芯片高通量表达谱分析及大规模微阵列技术,筛选鼻咽癌与炎症组织中差异表达的miRs,并应用RT-QPCR方法验证筛选结果的可信性。结果:鼻咽癌组织与对照组织的miRs存在差异表达,其中差异倍数大于2倍的人属miRs共有144种。RT-QPCR发现与炎症组织相比,癌组织中miRs-34b、miRs-449b、miRs-7-1表达显著下调,而miRs-125b、miRs-184、miRs-196b、miRs-205及miRs-24-1表达上调,结果与芯片分析结果相一致。结论:差异表达的miRs可能与鼻咽癌的发生、发展密切相关,对鼻咽癌miRs表达谱的研究可能为鼻咽癌的早期诊断及治疗提供有力的靶向依据。
[Abstract]:Objective: to screen and verify the differential expression profiles of microRN AsmiRsin nasopharyngeal carcinoma (NPC). Methods: the differentially expressed miRsin nasopharyngeal carcinoma (NPC) and inflammatory tissues were screened by high throughput microarray analysis and microarray technique. The RT-QPCR method was used to verify the credibility of the screening results. Results: there was a significant difference in miRs expression between nasopharyngeal carcinoma tissues and control tissues. There were 144 species of miRs in which the difference times were more than 2 times. RT-QPCR revealed that the expression of miRs-34bnmiRs-449brs-7-1 was significantly down-regulated, while miRs-125bnmiRs-184miRs-184miRs-196bnmiRs-205 and miRs-24-1 were up-regulated. The results are consistent with the results of chip analysis. Conclusion: differential expression of miRs may be closely related to the occurrence and development of nasopharyngeal carcinoma. The study of miRs expression profile of nasopharyngeal carcinoma may provide an effective target for the early diagnosis and treatment of nasopharyngeal carcinoma.
