去整合素Kistrin对晶状体摘除术后兔晶状体上皮细胞MMP-2和TIMP-2基因表达的影响

【摘要】:目的:研究去整合素Kistrin作用下,晶状体摘取术后兔晶状体上皮细胞( Lens epithelium cells , LECs )中基质金属蛋白酶2 ( Matrix metalloproteinases 2,MMP-2)mRNA和其抑制因子组织性金属蛋白酶抑制物2(Tissue inhibitor of metalloproteinase 2,TIMP-2)mRNA表达的变化,探讨kistrin作用的分子机制。 方法:将12只新西兰大白兔随机分为实验组和对照组,每组6只,均行右眼ECLE建立后V⒛せ熳莵\汀J醣鲜笛樽槟掖阆谧⑷肱ǘ任?0μg·L ~(-1)的Kistrin 0.2 mL,对照组注入等量生理盐水,随机选6只左眼为空白组。术后1 d、2d、3 d、7 d、14 d在裂隙灯下观察实验动物眼部情况。观察14天后取后V⒛ば惺凳庇舛縋CR检测MMP-2mRNA和TIMP-2mRNA的表达量。 结果:正常的兔后V⒛ど希ǹ瞻鬃椋㎝MP-2mRNA和TIMP-2mRNA表达量少,分别为0.127±0.039,2.485±0.507。白内障术后14天对照组两个基因表达量分别为3.785±0.934,19.903±10.499,均较空白组明显升高(p0.05)。实验组MMP-2mRNA的表达量为0.401±0.253,较对照组明显降低(p0.05),而与空白组比较无明显差异(p0.05)。实验组TIMP-2mRNA的表达量为16.119±3.773,与对照组比较无明显差异(p0.05),较空白组明显升高(p0.05)。 结论:晶状体囊外摘除术后使用Kistrin,MMP-2mRNA表达降低,TIMP-2mRNA的表达没有明显改变,可能是晶状体上皮细胞增殖受抑制的关键因素。
[Abstract]:Objective: to study the expression of matrix metalloproteinase 2 (Matrix metalloproteinases 2 mali-2md2 and tissue inhibitor of metalloproteinase (2(Tissue inhibitor of metalloproteinase 2) TIMP 2 in rabbit lens epithelial cells (LECs) after lens extraction (LECs) with disintegrin Kistrin, and to investigate the expression of MMP-2 mRNA and tissue inhibitor of metalloproteinase (2(Tissue inhibitor of metalloproteinase 2) in rabbit lens epithelial cells after lens extraction. To study the molecular mechanism of kistrin action. Methods: twelve New Zealand white rabbits were randomly divided into experimental group (n = 6) and control group (n = 6). After the establishment of right eye ECLE, Kistrin 0.2 mL was injected into the control group. Six left eyes were randomly selected as blank group. The eyes of experimental animals were observed under slit lamp at 1 day, 2 days, 3 days, 7 days and 14 days after operation. After 14 days of observation, V20 was removed. Ill-fated? The expression of MMP-2mRNA and TIMP-2mRNA was detected by lower CR. Results: normal rabbits after V20? The expression of MP-2mRNA and TIMP-2mRNA were 0.127 卤0.039 卤0.507 and 2.485 卤0.507 respectively. On the 14th day after cataract operation, the expression of two genes in the control group was 3.785 卤0.934 卤19.903 卤10.499, which was significantly higher than that in the blank group. The expression of MMP-2mRNA in the experimental group was 0.401 卤0.253, which was significantly lower than that in the control group, but there was no significant difference between the control group and the control group. The expression of TIMP-2mRNA in the experimental group was 16.119 卤3.773. There was no significant difference between the experimental group and the control group (P 0.05), but it was significantly higher than that in the blank group. Conclusion: after extracapsular lens excision, the expression of Kistrin MMP 2 mRNA decreased and TIMP-2 mRNA expression did not change significantly, which may be the key factor to inhibit the proliferation of lens epithelial cells.
