裸鼠喉癌细胞系Hep-2移植瘤中STK15基因的表达及意义

【摘要】: 实验目的 观察裸鼠喉癌细胞系Hep-2移植瘤的成瘤率,绘制肿瘤生长曲线,用免疫组化方法观察移植瘤中STK15基因的表达情况。 实验方法 将符合条件的17只nu/nu纯系裸小鼠随机分成两组,实验组10只,对照组7只。实验组每只裸鼠右侧前肢腋下皮下注射0.2mlHep-2细胞悬液,直至皮下形成0.5cm大小皮丘。对照组每只裸鼠相同部位注射0.2ml生理盐水,直至皮下形成0.5cm大小皮丘,定期观察两组裸鼠接种部位液体的吸收情况,计算成瘤率,绘制成瘤曲线。5周后处死实验动物,剥离肿瘤,用免疫组化方法观察移植瘤中STK15基因的表达情况。 实验结果 实验组裸鼠成瘤率100%,免疫组化可证实裸鼠喉癌细胞系Hep-2移植瘤中STK15过表达。 实验结论 通过动物实验证实STK15基因是喉癌的发生发展的原因之一。
[Abstract]:experimental purpose
The tumor growth curve was observed and the expression of STK15 gene in transplanted tumor was observed by immunohistochemistry .
experimental method
Two groups of nude mice were randomly divided into two groups , the experimental group was 10 rats and the control group was 7 . The experimental group injected 0.2 ml of normal saline at the same location on the right anterior limb of the nude mice , and then injected 0.2ml normal saline at the same site of the nude mice . The tumor rate was calculated and the tumor rate was calculated . After 5 weeks , the experimental animals were sacrificed and the tumor was stripped . The expression of STK15 gene was observed by immunohistochemistry .
experimental results
The tumor rate of nude mice in experimental group was 100 % , and the expression of STK15 was confirmed by immunohistochemistry .
experimental conclusion
Animal experiments prove that the STK15 gene is one of the causes of the development of laryngeal cancer .
