神经生长因子对猫双眼融合功能影响的探讨

【摘要】:目的: 从分子水平观察神经生长因子(nerve growth factor, NGF)在不同年龄正常猫融合中枢视皮层17区的表达量以探讨融合功能的发生机制及与其功能相关的可能因素。 方法: 正常2~3周健康家猫12只,应用图形视觉诱发电位(pattern visual evoked potential, PVEP)检测4周、6周、12周、16周幼猫双眼P波振幅和潜时,比较不同年龄P波及双眼间P波的差别。应用免疫组织化学方法比较不同年龄正常猫融合中枢视皮层17区NGF免疫阳性细胞量的差别,以判断NGF对双眼融合功能的影响。 结果: PVEP检测结果显示正常猫双眼间P波振幅和潜时之间均无统计学意义(P0.05),正常猫双眼间无明显差别。随着正常猫年龄的增长,P波振幅呈逐渐增加,潜时呈逐渐减少趋势。免疫组织化学方法观察视皮层17区阳性表达发现从4周龄开始皮层17区皮质Ⅱ层-Ⅳ层NGF免疫阳性细胞量逐渐增加,主要表现为颗粒细胞增多,6周时达到最大,从6周到12周逐渐下降,12周时降至接近但仍高于4周水平,16周降至明显低于4周时的表达水平。6周时表达量与4周时差异有高度统计学意义(P0.001),6周时与12周、16周NGF表达量差异均有高度统计学意义(P0.001),4周时与12周时NGF阳性细胞表达量差异有统计学意义(P0.05),与16周时NGF阳性细胞表达量差异有高度统计学意义(P0.001)。 结论: 随着正常猫年龄的增大,P波振幅逐渐增加,潜时逐渐减少。PVEP能够简单反映视功能的发育情况,说明随着年龄增长,视力也在不断发育的。根据各年龄正常幼猫视皮层17区Ⅱ层-Ⅳ层NGF表达量的差异可知,NGF与双眼融合功能的发育和形成有关。猫双眼融合功能随年龄增大逐渐发育,于视觉可塑性关键期6周左右发育最明显,之后呈逐渐下降趋势,NGF表达的变化与双眼视觉发育关键期保持高度一致,说明NGF是融合功能可塑性相关的重要分子基础。
[Abstract]:Objective: The expression of nerve growth factor (NGF) in the 17 area of fusion center of normal cats at different ages was observed at molecular level in order to explore the mechanism of fusion function and the possible factors related to its function. Methods: Visual evoked potential (VEP) pattern visual evoked potential, PVEP) was used to detect the amplitude and latent time of P wave in the eyes of young cats at 4 weeks, 6 weeks, 12 weeks and 16 weeks, and the differences of P waves between eyes of different ages were compared. Immunohistochemical method was used to compare the number of NGF immunoreactive cells in the 17 region of the fusion central visual cortex of normal cats of different ages, in order to determine the effect of NGF on binocular fusion function. Results: The results of PVEP showed that there was no significant difference in P wave amplitude and latent time between normal cats and normal cats, but there was no significant difference between normal cats and normal cats. The amplitude of P wave increased gradually and the latent time decreased with the increase of normal cat age.
