快速老化小鼠听功能及耳蜗组织神经生长因子受体酪氨酸激酶受体A的增龄性变化

【摘要】:目的:探讨快速老化小鼠听觉功能以及耳蜗组织中神经生长因子受体酪氨酸激酶受体A(nerve growth factor receptor TrkA,NGFR TrkA)表达的增龄性变化。方法 :选用3、5、7月龄的快速老化小鼠亚系8(SAMP8)作为观察对象。应用听觉诱发反应仪检测其8 kHz短纯音听觉脑干反应阈值;应用NGFR TrkA免疫组化染色检测其耳蜗组织染色光密度进行分析。结果:SAMP8听性脑干反应阈值随着月龄增加而显著增高(P0.05);NGFR TrkA蛋白在不同月龄快速老化小鼠耳蜗组织中均有表达,SAMP8耳蜗组织中的NGFR TrkA蛋白染色光密度随着月龄增加而显著降低(P0.05)。结论:NGFR TrkA蛋白的表达水平随着快速老化小鼠听觉功能增龄性减退而降低,说明NGFR TrkA可能与维持耳蜗的功能状态有关。
[Abstract]:Aim: to investigate the aging changes of auditory function and expression of nerve growth factor receptor tyrosine kinase receptor (A(nerve growth factor receptor TrkANGFR) in cochlea of rapidly aging mice. Methods: the rapid aging mouse subline 8 (SAMP8), aged from 3 to 5 to 7 months old, was selected as the observation object. Auditory evoked response apparatus was used to detect the auditory brainstem response threshold of 8 kHz short tone auditory brainstem, and NGFR TrkA immunohistochemical staining was used to detect the optical density of the cochlea tissue staining. Results the threshold of auditory brainstem response of SAMP8 was significantly increased with the increase of age. The expression of P0.05 TrkA protein in cochlea of different months of aging mice was significantly increased. The light density of NGFR TrkA protein in the cochlea of SAMP8 cochlea decreased significantly with the increase of age. Conclusion the expression level of TrkA protein decreases with the aging of auditory function in rapidly aging mice, suggesting that NGFR TrkA may be related to maintaining the functional state of cochlea.
