口服米诺环素对兔慢性高眼压模型的视神经保护作用

【摘要】: 目的:探索口服米诺环素胶囊(minocycline)对兔慢性高眼压模型视神经损伤的保护作用,并探讨其机制。 方法:取健康成年家兔48只,随机分为A组(正常组)、B组(高眼压组)和C组(高眼压治疗组),每组16只。B组和C组每只兔左眼为正常眼,右眼前房内注射3g/L复方卡波姆溶液0.1mL,造慢性高眼压模型。实验以各组兔右眼为观察对象。造模成功后,C组兔连续28d口服米诺环素胶囊,50mg/kg/24h。每周相同时间点测B组和C组兔眼压,并记录数据。第29d用耳缘静脉注气法处死各组动物,三组各随机取8只兔,剥取右眼视网膜,制成100g/L匀浆液。考马斯亮蓝染色法(Commassie Blue Staining Kit)测出匀浆蛋白含量后以比色法测SOD、MDA、NO,并计算出各指标含量。每组另外8个眼球进行常规固定、脱水、石蜡包埋切片,免疫组化法测caspase-3含量,阳性结果以胞浆或胞核内出现棕黄色染色为准。Image-Pro Plus5.0图像分析软件检测每个标本所测量的面积内,组织的平均灰度值(IOD)。SPSS13.0统计软件分析处理每周相同时间点眼压、超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)含量、caspase-3平均灰度值,并进行组间比较分析。 结果:B组和C组右眼比较,每周相同时间点眼压测量结果无显著统计学差异,均大于22mmHg。 caspase-3免疫组化检测结果显示,A组兔观察不到棕黄色的阳性染色,视网膜caspase-3平均灰度值极低;B,C两组可以观察到相应阳性染色颗粒。B组与A组比较,平均灰度值均较高,差异具有统计学意义;C组视网膜组织Caspase-3平均灰度值明显低于B组,差异具有统计学意义;A组视网膜组织Caspase-3平均灰度值低于C组,差异具有统计学意义。 兔视网膜匀浆中SOD、NO、MDA表达测定结果显示:与A组相比,B组SOD值明显较低,NO、MDA较高,差异具有统计学意义;C组与B组相比,SOD值明显较高;NO、MDA较低,差异具有统计学意义;C组SOD值明显低于A组;NO、MDA明显高于A组,差异具有统计学意义。 结论:兔眼前房内注射复方卡波姆溶液,诱导出的慢性高眼压模型观察的时间内眼压可以维持在较高水平。口服一定剂量的米诺环素胶囊对慢性高眼压下兔视网膜神经节细胞的损伤有一定保护作用,其潜在作用机制可能是它所具有的抗凋亡、抗氧化的作用,但这种保护机制与眼压无关。
[Abstract]:Aim: to investigate the protective effect of minocycline capsule on optic nerve injury in rabbits with chronic intraocular hypertension and its mechanism. Methods: Forty-eight healthy adult rabbits were randomly divided into two groups: group A (normal group) and group C (high intraocular pressure group), with 16 rabbits in each group and 16 rabbits in group C as normal eyes. The model of chronic high intraocular pressure was established by injecting 3g/L compound carbomer solution (0.1 mL) into right anterior chamber. The right eye of each group was observed. After the successful establishment of the model, the rabbits in group C took minocycline capsule 50 mg / kg / 24 h for 28 days. Intraocular pressure (IOP) of group B and group C were measured and recorded at the same time every week. On the 29th day, the animals in each group were killed by auricular vein injection. Eight rabbits were randomly selected from each group, and the right eye retina was stripped to make 100g/L homogenate. Coomassie brilliant blue staining method and Commassie Blue Staining Kit) were used to determine the content of homogenate protein, and then to measure the content of SDD MDAN by colorimetric method, and to calculate the contents of each index. The other 8 eyeballs in each group were routinely fixed, dehydrated, embedded in paraffin sections, and the content of caspase-3 was measured by immunohistochemical method. The positive results were detected by the brown-yellow staining in cytoplasm or nucleus. Image-Pro Plus5.0 image analysis software was used to detect the area of each specimen. The mean gray value of tissue was analyzed by IODX. SPSS 13.0 statistical software. IOP, SOD, MDA, no) content and caspase-3 mean gray value were analyzed at the same time point every week. Results there was no significant difference in intraocular pressure between group B and group C at the same time point every week, all of which were more than 22mm Hg. The results of caspase-3 immunohistochemical staining showed that the positive staining of brown was not observed in group A, and the average gray value of caspase-3 in group B was higher than that in group A, and that in group B was significantly higher than that in group A. The average gray value of Caspase-3 in group C was significantly lower than that in group B, and the average gray value of Caspase-3 in group A was lower than that in group C. the difference was statistically significant. The expression of SOD in rabbit retina homogenate was significantly lower than that in group A (P < 0.05). The difference was statistically significant between group C and group B (P < 0.05), and the level of SOD in group C was significantly higher than that in group B (P < 0.05). The SOD of group C was significantly lower than that of group A, and the difference was statistically significant. Conclusion: the intraocular pressure induced by the injection of compound carbomer solution into rabbit anterior chamber can be maintained at a high level during the observation period. A certain dose of minocycline capsule could protect the retinal ganglion cells from injury induced by chronic intraocular pressure, and its potential mechanism might be its anti-apoptosis and anti-oxidation effects. But this protective mechanism has nothing to do with intraocular pressure.
