激光光凝小梁网建立大鼠青光眼模型眼压变化规律的研究

【摘要】:目的:探讨532-二极管激光建立大鼠青光眼模型眼压的变化规律。 方法:分别选用健康2月龄和4月龄Wistar大鼠各40只,每组光凝30只,其余10只作空白对照观测眼压。光凝一眼,另一眼为自身对照,左右眼交替。每7天光凝一次,共4次,满1月时存活的大鼠随机选择一半作为实验组,和空白对照组同时在全麻下取出双眼眼球;另一半为继续观察组,再继续观测眼压1月,满2月时在全麻下取出双眼眼球。光凝方法:采用532-二极管激光经角膜缘光凝小梁网360°,激光参数设置为0.2s/0.5w,每只眼光凝约150个点。分别于光凝前、光凝后即刻及光凝后每天同一时间段用Tonolab眼压计测量大鼠双眼眼压,每只眼连续测量6次,取平均值,同时观察双眼前节情况。 结果:2月龄大鼠第一次光凝前平均眼压为15±3.8mmHg,光凝后即刻平均眼压为24±6.3mmHg,差异有统计学意义(t=6.632,P0.001);4月龄大鼠第一次光凝前平均眼压为13±2.8mmHg,光凝后即刻平均眼压为26±10.8mmHg,差异有统计学意义(t=6.485, P0.001);光凝后即刻2月龄和4月龄大鼠眼压峰值分别为60mmHg和54mmHg。两组大鼠光凝后即刻平均眼压差异无统计学意义(t=-1.623, P0.05)。光凝后第1天平均眼压最高然后逐日下降,在第6天恢复到正常范围。在光凝后6天观测中,2月龄和4月龄眼压峰值分别为66 mmHg和74mmHg。两组大鼠每次重复光凝后眼压均有上述相同的变化。2月龄第一次光凝后即刻眼压升高的动物为63%,4月龄为60%。眼压升高动物最多出现在光凝后第1天,2月龄可达到77%,4月龄可达到88%,均出现在第四次光凝后的第1天。大多数动物眼压在光凝后第6天恢复到正常范围。 结论:532-二极管激光经角膜缘光凝小梁网建立的大鼠青光眼模型有可能是一过性眼压升高,大鼠慢性青光眼动物模型可能需反复多次光凝小梁网才能获得。
[Abstract]:Objective: to investigate the changes of intraocular pressure (IOP) in rat model of glaucoma induced by 532-diode laser. Methods: 40 healthy Wistar rats aged 2 months and 4 months were selected, 30 rats in each group were photocoagulated, the other 10 rats were used as blank control to observe intraocular pressure (IOP). Light coagulation, another eye for self-control, left and right eyes alternately. Once every 7 days, 4 times a day, half of the rats who survived at the end of one month were randomly selected as the experimental group, and the eyes of both eyes were taken out under general anesthesia with the blank control group. The other half was the continuous observation group, and then the IOP was observed for 1 month. Take out your eyes under general anesthesia for two months. The method of photocoagulation: using 532-diode laser through the limbal photocoagulation trabecular net 360 掳, the laser parameter is set to 0.2 s / 0.5 w.Each eye sets about 150 points. The intraocular pressure (IOP) was measured by Tonolab intraocular pressure meter at the same time of day before photocoagulation, immediately after photocoagulation and at the same time after photocoagulation. The intraocular pressure of each eye was measured for 6 consecutive times, and the mean value was taken, and the anterior segment of the eyes was observed at the same time. Results the mean IOP was 15 卤3.8 mmHg before the first photocoagulation and 24 卤6.3 mmHg immediately after photocoagulation in 2 month old rats. The difference was statistically significant (13 卤2.8 mmHgand 26 卤10.8mmHg respectively). The peak IOP of 2-month and 4-month-old rats was 60mmHg and 54mm Hg, respectively. There was no significant difference in IOP between the two groups immediately after photocoagulation (P 0.05). The mean IOP was highest on the 1st day after photocoagulation, then decreased daily, and returned to normal on the 6th day. The peak values of IOP at 2 and 4 months of age were 66 mmHg and 74 mm Hg, respectively, 6 days after photocoagulation. The intraocular pressure (IOP) of the rats in the two groups increased immediately after photocoagulation for the first time at the age of 2 months, and the IOP was 60 months old at the age of 63 to 4 months after repeated photocoagulation. The elevation of intraocular pressure (IOP) occurred mostly on the 1st day and 2nd month after photocoagulation, and the age of 77 and 4 months could reach 888.All of them appeared on the first day after the fourth photocoagulation. IOP in most animals returns to normal on the 6th day after photocoagulation. Conclusion the rat model of glaucoma induced by 532 diode laser through the limbal photocoagulation trabecular reticulum may be a transient elevation of intraocular pressure, and the animal model of chronic glaucoma may be obtained by repeated photocoagulation of trabecular meshwork.
