HSP90和eNOS在激素性青光眼发病中的作用

【摘要】:目的 探讨HSP90和内皮型一氧化氮合酶(eNOS)在糖皮质激素性青光眼发病中的作用,以及与视网膜神经节细胞的凋亡之间的内在联系。为今后对糖皮质激素性青光眼的进一步研究和治疗提供理论依据。 方法 28只新西兰大白兔,随机分成4组,对照组、实验A组、B组、C组,每组7只,实验组隔日定时左眼球结膜下注射地塞米松0.5mg(0.1ml)。对照组隔日左眼球结膜下注射等量生理盐水。各组隔日定时测左眼眼压。A组用药两周后进行指标检测,B组和C组继续隔日定时左眼球结膜下注射地塞米松0.5mg(0.1ml)共四周。四周后B组进行各项指标测定,C组停药观察眼压变化后进行指标测定。免疫组化观察每组小梁网中eNOS的阳性表达和视网膜中HSP90的阳性表达,TUNEL法检测视网膜神经节细胞(RGCs)的凋亡情况。 结果 正常对照组的眼压在各个时间段变化差别无统计学意义(P0.05)。模型组中用药4次后眼压开始升高,随着用药时间的延长眼压升高明显。A、B、C组用药两周后眼压升高明显,与对照组比较差别有统计学意义(P0.05)。实验组B组和C组用药四周后眼压升高到实验的峰值,与对照组比较差异性显著(P0.01)。视网膜中的HSP90阳性表达和小梁网中的eNOS阳性表达随着用药时间的延长与对照组比较阳性表达明显减少,A组与对照组比较差别有统计学意义(P0.05),B、C组与对照组比较差异性显著(P0.01)。A、B、C组RGCs细胞随着用药时间的延长眼压的增高凋亡逐渐增多。 结论 HSP90和eNOS的阳性表达都与视网膜神经节细胞的凋亡之间存在显著的负相关。充分说明两者的含量减少在糖皮质激素性青光眼发病中起着至关重要的作用,为今后对糖皮质激素性青光眼的进一步研究和治疗提供理论依据。
[Abstract]:Purpose To investigate the role of HSP90 and endothelial nitric oxide synthase (Enos) in the pathogenesis of glucocorticoid glaucoma and the relationship between them and the apoptosis of retinal ganglion cells. To provide theoretical basis for the further study and treatment of glucocorticoid glaucoma. Method Twenty-eight New Zealand white rabbits were randomly divided into 4 groups: control group and experimental group A and B with 7 rabbits in each group. The experimental group was injected with dexamethasone 0.5 mg / L 0.1 ml / L subconjunctival dexamethasone every other day. In the control group, the same amount of saline was injected under the bulbar conjunctiva of the left eye every other day. Left intraocular pressure (IOP) in group A was measured two weeks later. The indexes of group B and group C continued to be measured by subconjunctival injection of dexamethasone (0.5 mg / ml) for four weeks. Four weeks later, the indexes were measured in group B and group C after stopping drug and observing the change of intraocular pressure. The expression of eNOS in trabecular meshwork and the expression of HSP90 in retina were observed by immunohistochemical staining and Tunel method was used to detect the apoptosis of retinal ganglion cells (RGCs). Result There was no significant difference in intraocular pressure (IOP) in normal control group (P 0.05). The IOP in the model group began to increase after 4 times of medication, and the IOP increased significantly with the prolongation of the time of administration. The IOP in group A BX C increased significantly after two weeks of treatment, and there was a significant difference between the two groups (P 0.05). The intraocular pressure in group B and group C increased to the experimental peak after four weeks, and the difference was significant compared with the control group (P 0.01). HSP90 positive expression in retina and eNOS positive expression in trabecular meshwork were significantly decreased compared with control group. There was significant difference between group A and control group (P 0.05). The apoptosis of RGCs cells increased with the prolongation of intraocular pressure (IOP). Conclusion There was a significant negative correlation between the positive expression of HSP90 and eNOS and the apoptosis of retinal ganglion cells. It is fully explained that the decrease of the content of both plays an important role in the pathogenesis of glucocorticoid glaucoma and provides a theoretical basis for the further study and treatment of glucocorticoid glaucoma in the future.
