Importin13在角膜缘肿瘤中的差异性表达

【摘要】：目的：研究一个新近发现的细胞核质双向转运受体蛋白importin13(IPO13)在人角结膜良、恶性肿瘤中的表达,并与角膜缘干细胞标记物p63作比较,探讨两者在人角结膜良、恶性肿瘤中的表达差异及意义。方法：实验分为三组：正常组6例,取自正常人眼球角膜缘处的结膜及角膜缘组织(来源于角膜移植术后的供体眼球)；角结膜乳头状瘤组(Conjunciva-cornea papilloma, CCP)6例、角结膜上皮内上皮癌组(Conjunctiva-cornea intraepithelial neoplasia, CCIN)6例,肿块标本均取自在湘雅医院眼科住院部接受手术治疗并经病理检查确诊的患者。采用冰冻切片免疫组织化学染色法检测IPO13、p63在人正常角结膜、CCP和CCIN三组角结膜组织中的差异性表达。结果：1.IP013表达于人正常眼球结膜、角膜缘上皮基底细胞的细胞核内,表达于CCP、CCIN组织上皮基底细胞和基底上细胞的细胞核内,并且在正常组、CCP、CCIN组织中的表达逐步上调,三组之间差异具有统计学意义(p0.05)。 2.p63表达于人正常眼球结膜、角膜缘、CCP组织上皮基底细胞和基底上细胞的细胞核内,在CCIN组织上皮全层的细胞核内均有表达,并且在正常组、CCP、CCIN组织中的表达逐步上调,三组之间差异具有统计学意义(p0.05)。结论：1.IP013在标志角膜缘上皮增殖性病变中的低分化细胞较p63更具特异性。 2.与正常组相比,IP013与p63在CCP、CCIN组织中的表达逐步上调,推测IPO13与p63可能在角结膜增殖性病变中起到正性调控作用。 3.IPO13与p63在良性与恶性角膜缘上皮增殖性病变中的表达差异明显,提示IP013与p63可能在角膜缘上皮增殖性病变的异常增殖与分化中起到重要调控作用。
[Abstract]:Objective: to study the expression of a recently discovered nuclear cytoplasmic bidirectional transport receptor protein import in13 (IPO13) in human keratoconjunctiva benign and malignant tumors, and to compare it with corneal limbal stem cell marker p63, and to investigate the expression of the two proteins in human keratoconjunctiva. Expression difference and significance in malignant tumor. Methods: the experiment was divided into three groups: normal group (n = 6), conjunctival tissue and limbal tissue (from donor eyeball after keratoplasty), conjunctival papillomia (CCP)6), conjunctival papilloma group (n = 6), and conjunctival papillomia group (n = 6). Conjunctiva-cornea intraepithelial neoplasia, CCIN)6, were collected from patients who underwent surgical treatment and confirmed by pathological examination in the ophthalmic hospital of Xiangya Hospital. Immunohistochemical staining of frozen sections was used to detect the differential expression of IPO13 p63 in the conjunctiva of human normal conjunctiva and CCIN. Results 1. IP013 was expressed in the nucleus of the basal cells of corneal limbal epithelium, in the nucleus of epithelial basal cells of CCPCCIN, and in the nuclei of epithelial basal cells and suprabasal cells of CCPCCIN, and the expression of IP013 was gradually up-regulated in the normal group of conjunctival conjunctiva and corneal limbal epithelial basal cells. The difference between the three groups was statistically significant (P 0.05). The expression of 2.p63 was found in the nucleus of basal cells and suprabasal cells of normal conjunctiva, limbal keratoconjunctiva, and the whole layer of CCIN tissue, and increased gradually in the normal group. The difference between the three groups was statistically significant (P 0.05). Conclusion: 1. The low differentiation cells of IP013 in the marker of limbal epithelial proliferative lesions are more specific than p63. 2. Compared with the normal group, the expression of p63 and IP013 was gradually up-regulated in CCIN, suggesting that IPO13 and p63 might play a positive role in the regulation of conjunctival proliferative lesions. The expression of 3.IPO13 and p63 in benign and malignant corneal limbal epithelial proliferative lesions was significantly different, suggesting that IP013 and p63 might play an important role in the abnormal proliferation and differentiation of corneal limbal epithelial proliferative lesions.
