支配眼部三叉神经神经元CGRP表达的实验研究

【摘要】:目的:观察大鼠三叉神经节内降钙素基因相关肽(Calcitonin gene-related peptide CGRP)阳性细胞的分布情况,及其对眼部组织的支配情况,初步了解CGRP与眼部痛觉产生及调控的相关性。 方法:随机抽取5只SD大鼠,10只眼,随机分为生理盐水对照组,上眼睑+球结膜注射组,球结膜注射组,上眼睑注射组,前房注射组。将生理盐水,荧光金分组注入大鼠球结膜,上眼睑及前房,逆行示踪三叉神经节神经元,同时用CGRP免疫荧光组织化学标记大鼠三叉神经节组织。在荧光显微镜下统计分析荧光金标记、CGRP免疫荧光分别标记及双重标记的三叉神经节神经元数量。采用SPSS 13.0统计软件对所得数据进行t检验。 结果:通过荧光金逆行示踪的方法,球结膜,上眼睑及前房分别注射组均发现在三叉神经节内可见荧光金标记的神经元,但在数量上存在一定的差异;运用免疫荧光组织化学染色的方法,三叉神经节内可见许多CGRP阳性神经元;两种方法相结合可以观察到少量荧光金标记的神经元同时也呈CGRP免疫荧光阳性。对标记的神经元计数,进行统计学分析,支配眼内组织感觉的神经元数量与支配球结膜及上眼睑的数量进行对比,P0.05,差异有统计学意义。 结论:三叉神经节内有大量CGRP免疫阳性神经元支配眼部组织,其中有少量神经元支配眼内组织,CGRP参与眼部伤害性刺激的调控。
[Abstract]:Objective: To observe the distribution of Calcitonin gene-related peptide CGRP positive cells in the trigeminal ganglion of the rat trigeminal ganglia, and the control of the ocular tissue, and to understand the correlation between CGRP and the production and regulation of ocular pain.
Methods: 5 SD rats were randomly selected, 10 eyes were randomly divided into normal saline control group, upper eyelid + conjunctival injection group, ball conjunctiva injection group, upper eyelid injection group and anterior chamber injection group. The saline and fluorescent gold groups were injected into the rat conjunctiva, upper eyelid and anterior chamber, and the trigeminal ganglion neurons were traced by inverse line, and CGRP immunofluorescence tissue was used. The trigeminal ganglion tissue of rats was marked by chemical labeling. The fluorescent gold labeling, CGRP immunofluorescence and the number of double labeled trigeminal ganglion neurons were statistically analyzed under the fluorescence microscope. The data were tested by the SPSS 13 statistical software for the t test.
Results: by the method of retrograde tracing of fluorescent gold, the neurons in the trigeminal ganglia were found in the conjunctiva, the upper eyelids and the anterior chamber respectively, but there were some differences in the number of neurons. Many CGRP positive neurons were seen in the trigeminal ganglion by immunofluorescence staining, and the two methods were found in the trigeminal ganglion. A small amount of fluorescent gold labeled neurons were also observed to be immunofluorescent with CGRP immunofluorescence. The count of labeled neurons was statistically analyzed, and the number of neurons that dominate the sensory tissue of the eye was compared with the amount of the domination of the conjunctiva and the upper eyelids, and the difference was statistically significant in P0.05.
Conclusion: there are a large number of CGRP immunoreactive neurons in the trigeminal ganglia, which dominate the ocular tissue, and a small number of neurons dominate the intraocular tissue, and CGRP is involved in the regulation of ocular nociceptive stimulation.
