水通道蛋白5与低氧反应通路因子(HIF-1α、VEGF)在鼻息肉组织中的表达及其相关性研究

【摘要】:目的:检测水通道蛋白5(aquaporin-5 , AQP5)、缺氧诱导因子1α( hypoxia-inducible factor-1α, HIF-1α)和血管内皮生长因子( vascular endothelial growth factor, VEGF)在鼻息肉组织中的表达情况,并探讨三种因子之间表达的相关性及其在鼻息肉发病机制中的作用。 方法:取18例患者鼻息肉组织及10例单纯行鼻中隔偏曲矫正术中切除的下鼻甲组织作为对照组,分别运用Real-time PCR (Real-time Quantitative Polymerase chain Reaction, RT-PCR)和蛋白免疫印迹(Western blot)检测AQP5、HIF-1α、VEGF mRNA以及蛋白的表达情况;采用t检验和直线相关分析进行统计学分析。 结果:①RT-PCR结果显示, AQP5mRNA在鼻息肉中的表达明显低于下鼻甲组织,差异有统计学意义( P 0.01) , HIF-1α、VEGF mRNA在鼻息肉和下鼻甲组织中表达差异无统计学意义( P 0.05) ;②Western blot结果显示AQP5蛋白表达鼻息肉组与对照组无明显差异。(P 0.05),HIF-1、VEGF蛋白表达水平在鼻息肉组织中表达显著高于对照组(P0.05);③Western blot结果显示鼻息肉组织中AQP5与HIF-1α之间蛋白的表达呈明显正相关关系(r =0.633, P 0.01), AQP5与VEGF蛋白质表达亦呈明显正相关关系(r =0.611, P 0.01)。 结论: AQP5与HIF-1α、VEGF等低氧反应通路因子共同参与鼻息肉组织水肿的形成。前者通过组织分布异常引起水转运障碍进而导致组织水肿形成,后者主要通过诱发血管增殖、增加血管通透性进而导致组织水肿产生;两者是相对独立的系统,我们推测它们可能通过某种间接途径发生联系。
[Abstract]:Objective: to investigate the expression of aquaporin-5 (AQP5), hypoxia-inducible factor-1 伪 (HIF-1 伪) and vascular endothelial growth factor (vascular endothelial growth factor,) in nasal polyps. The relationship between the expression of three factors and its role in the pathogenesis of nasal polyps was discussed. Methods: Real-time Polymerase chain Reactionation (RT-PCR) and Western blot (Western blot) were used to detect the expression of AQP5, HIF-1 伪 -VEGF mRNA and protein in 18 cases of nasal polyps and 10 cases of inferior turbinate tissue removed during nasal septum deviation correction. T test and linear correlation analysis were used for statistical analysis. Results the expression of AQP5 mRNA in nasal polyps was significantly lower than that in inferior turbinate tissues (P0.01), but there was no significant difference in HIF-1 伪 -VEGF mRNA expression in nasal polyps and inferior turbinate tissues (P 0.05). 2Western blot showed that the expression of AQP5 protein in nasal polyp group was not significantly different from that in control group. (P 0.05) the expression of HIF-1 protein in nasal polyp tissue was significantly higher than that in control group (P0.05). Western blot showed that AQP5 and HIF-1 伪 in nasal polyp tissue were between AQP5 and HIF-1 伪. The expression of white was positively correlated with that of VEGF (r = 0.633, P 0.01), and the expression of AQP5 was also positively correlated with the expression of VEGF (r = 0.611, P 0.01). Conclusion: AQP5 and HIF-1 伪 -VEGF are involved in the formation of edema in nasal polyps. The former leads to water transport disturbance through abnormal tissue distribution, which leads to the formation of tissue edema, while the latter mainly induces vascular proliferation, increases vascular permeability and leads to tissue edema, which is a relatively independent system. We speculate that they may be connected in some indirect way.
