人葡萄膜黑色素瘤小鼠肝内种植模型生长行为研究
[Abstract]:Aim: to study the biological behavior of human uveal malignant melanoma cell line (MUM2B) in vivo by intrahepatic injection of human uveal malignant melanoma strain (MUM2B cell). Methods: 5 脳 104 MUM2B cells were injected into the liver capsule of BALB/C-nu nude mice. For 6 weeks, 3 nude mice were randomly killed to observe the tumorigenesis rate and invasion rate. The tumor bearing liver, heart, spleen, lung, kidney, stomach and intestine of nude mice were collected. The tumor growth behavior in liver and its invasion and metastasis to other organs were examined by HE staining. Results: the tumorigenesis rate of the model was 83.3%. In the first 4 weeks after inoculation, the tumor grew infiltrating in the liver and did not invade the surrounding organs. At the 5th week after inoculation, the tumor began to invade the capsule of kidney, spleen, stomach and duodenum, and it could break through the capsule of stomach and intestine and invade and grow in its serous layer. At 6 weeks after inoculation, the tumor increased invasion of kidney, spleen, duodenum and stomach. Tumor cells could infiltrate the myometrium of duodenum of nude mice, invade the gastric myometrium and submucosa of nude mice, and even invade into the intramuscular layer of gastric mucosa. At the 6th week after inoculation, the tumor began to metastasize to the lung of nude mice, and metastasis could be found in hilar and parenchyma of all the tested nude mice, and the metastatic rate was 100%. No metastatic tumor foci were found in the other organs except the lung suggesting that the lung was the main metastatic target organ of the implanted tumor in the liver. In addition, after 2 weeks of bearing tumor, the proliferation of red myeloid in spleen of nude mice was obvious. A large number of megakaryocytes and myeloid granulocytes could be seen in the marrow of nude mice, showing leukaemia like reaction. After 4 weeks of bearing tumor, the star-like phenomenon in spleen was obvious and macrophage was active, suggesting that the histological changes of spleen in nude mice were closely related to the growth state of bearing tumor. Conclusion: intrahepatic injection of MUM2B cells in nude mice can establish a stable intrahepatic implantation model of human uveal melanoma mice. The implanted tumor cells infiltrate the liver of nude mice, invading many organs, including stomach, duodenum, spleen and kidney, and metastasis to the lung.
