α-硫辛酸对小鼠视网膜光化学损伤的保护作用

【摘要】:目的观察α-硫辛酸对光损伤后小鼠视网膜的保护作用。方法将42只成年健康BALB/C小鼠随机分为正常组、光损伤组和药物干预组(干预组又分为造模前后均给药组及造模后给药组)。除正常组外其他组小鼠均暴露于装有白色光源的光损伤箱内,光暴露时间为12h,之后暗室饲养12h,共3个循环,光照强度21400±1600 lx,总暴露时间为36h。箱内温度控制在25-29℃。药物干预组腹腔注射α-硫辛酸注射液100mg/Kg,一次/日,直至处死。分别于7d及14 d处死动物摘取眼球,制备眼球壁石蜡切片,在光学显微镜下观察其形态学变化,,测量视网膜外核层厚度,用TUNEL法计数外核层细胞凋亡数及凋亡指数,用x±s表示,各组之间比较用两因素析因设计的方差分析。结果与光损伤组相比,药物干预组视网膜结构层次分明,细胞排列整齐。其中视网膜外核层厚度,正常组41.62±0.77um,光损伤组7天30.51±0.73um,14天20.04±0.52um,药物干预组(C1组)7天40.52±0.80um,14天41.43±0.72um,造模后给药组(C2组)7天31.70±0.0.70um,14天38.92±1.43um,显著高于光损伤组,(p0.001)。Tunel试剂盒检测细胞凋亡结果显示:正常组AI1,光损伤组AI=15.44±1.72,药物干预Ⅰ组AI=7.23±0.65,药物干预Ⅱ组AI=7.48±0.88。给药组小鼠视网膜外核层的凋亡细胞数明显少于光损伤组小鼠。外核层(ONL)厚度及凋亡指数(AI)的组间差异具有统计学意义。结论在本实验条件下,α-硫辛酸对小鼠视网膜光损伤具有保护作用。
[Abstract]:Objective to observe the protective effect of 伪-lipoic acid on the retina of mice after light injury. Methods 42 healthy adult BALB/C mice were randomly divided into three groups: normal group, light injury group and drug intervention group.
