膜型1基质金属蛋白酶(MT1-MMP)在视网膜前膜中的表达

【摘要】:研究目的: 1.研究膜型1基质金属蛋白酶(MT1-MMP)在不同视网膜前膜(ERM)组织中的表达差异。 2.研究MT1-MMP在视网膜前膜新生血管内皮细胞中的表达及分布情况。 研究方法: 1.收集增殖性糖尿病视网膜病变(PDR),增殖性玻璃体视网膜病变(PVR)及特发性黄斑前膜(IERMM)患者的ERM标本及临床资料。 2.通过免疫组织化学染色,观察MT1-MMP在不同ERM中的表达强弱,并通过图像分析软件(Image Pro Plus6.0)分析其累计光密度(IOD)值。 3.通过免疫荧光双重标记染色,以CD34标记ERM中新生血管内皮细胞,观察MT1-MMP在内皮细胞中的表达及分布。 研究结果: 1.共收集33例ERM标本,其中PDR患者22例,非PDR患者11例(PVR患者7例,IERMM4例)。 2.在PDR和非PDR患者的ERM标本中,MT1-MMP表达阳性率分别为77.3%(17/22)和18.2%(2/11)(p=0.002);在PDR和非PDR患者的ERM标本中,MT1-MMP表达的平均IOD值分别为5767.36±2505.03和1728.73±717.32(p=0.000)。 3.在PDR患者的ERM标本中,MT1-MMP与CD34免疫荧光染色阳性,分布较一致,均位于细胞膜上。在非PDR患者的ERM标本中,MT1-MMP与CD34免疫荧光染色阴性。 研究结论: 1.与对照组相比,糖尿病视网膜病变视网膜前膜显著高表达MT1-MMP, MT1-MMP可能在糖尿病视网膜病变增殖膜形成中发挥一定作用。 2.糖尿病视网膜病变的视网膜前膜中含有较多新生血管成分,MT1-MMP表达在糖尿病视网膜病变视网膜前膜新生血管内皮细胞的细胞膜上,MT1-MMP可能在糖尿病视网膜病变视网膜前膜的新生血管形成中发挥一定作用。
[Abstract]:Research purposes: 1. To study the expression of membrane type 1 matrix metalloproteinases (MT1-MMP) in different (ERM) tissues. 2. To study the expression and distribution of MT1-MMP in endothelial cells of epiretinal neovascularization. Methods of research: 1. ERM specimens and clinical data of patients with proliferative diabetic retinopathy (PDR), proliferative vitreoretinopathy (PVR) and idiopathic macular anterior membrane (IERMM) were collected. 2. The expression of MT1-MMP in different ERM was observed by immunohistochemical staining, and the cumulative optical density (IOD) was analyzed by image analysis software (Image Pro Plus6.0). 3. The expression and distribution of MT1-MMP in ERM were observed by double immunofluorescence staining, and the expression and distribution of MT1-MMP in endothelial cells were observed by using CD34 to label the neovascularization endothelial cells. Research findings: 1. 33 cases of ERM were collected, including 22 cases of PDR and 11 cases of non-PDR (7 cases of PVR and 4 cases of IERMM). 2. The positive rates of MT1-MMP expression in ERM samples from PDR and non-PDR patients were 77.3% (17 / 22) and 18.2% (2 / 11) respectively (p = 0.002). In ERM specimens from PDR and non-PDR patients, the average IOD values of MT1-MMP expression were 5767.36 卤2505.03 and 1728.73 卤717.32 (p < 0. 000), respectively. 3. In the ERM samples of PDR patients, MT1-MMP and CD34 immunofluorescence staining were positive, the distribution was consistent, both of them were located on the cell membrane. MT1-MMP and CD34 immunofluorescence staining was negative in ERM samples from non-PDR patients. Research conclusions: 1. Compared with the control group, the high expression of MT1-MMP, MT1-MMP in the epiretinal membrane of diabetic retinopathy may play a role in the formation of proliferative membrane in diabetic retinopathy. 2. The epiretinal membrane of diabetic retinopathy contains more neovascularization components, and MT1-MMP is expressed on the membrane of neovascularization endothelial cells in diabetic retinopathy. MT1-MMP may play a role in the neovascularization of diabetic retinopathy.
