兔眼脉络膜撕脱伤后bFGF与PDGF-A的表达变化

【摘要】:目的:建立兔眼脉络膜撕脱伤模型,观察不同时间点碱性成纤维细胞生长因子(basic fibroblast growth factor,bFGF)与血小板源性生长因子(platelet derived growth factor-A,PDGF-A)在脉络膜、视网膜及玻璃体内的表达变化,探讨脉络膜撕脱伤后bFGF与PDGF-A在脉络膜增殖修复中作用,为临床治疗提供新的理论依据。 方法:40只成年白兔随机分成5组,包括正常组,伤后3d、7d、14d、28d实验组,每组各8只,奇偶数法确定实验眼;建立兔眼脉络膜撕脱伤模型,于伤后不同时间点观察眼底变化并行眼科B超检查;HE染色光镜下观察伤口处脉络膜、视网膜形态学变化;免疫组化SP法和酶联免疫吸附实验(Enzyme-Linked Immunosorbnent Assay,ELISA)方法分别检测脉络膜、视网膜及玻璃体内bFGF与PDGF-A蛋白表达量的变化。 结果:伤后脉络膜、视网膜及玻璃体内bFGF与PDGF-A表达上调。bFGF与PDGF-A蛋白在脉络膜、视网膜内表达的平均光密度值及在玻璃体内表达的平均OD值均出现先升高又下降的变化,于伤后3d明显升高,7d达到高峰,14d略有回落,仍维持较高水平,28d明显下降,但高于正常。正常组、各实验组比较,差异有统计学意义(P0.05);伤后7d与14d比较,差异无统计学意义(P0.05),伤后7d分别与伤后3d、28d比较,差异有统计学意义(P0.05)。 结论:①bFGF与PDGF-A参与脉络膜撕脱伤后脉络膜的增殖修复的发生发展,是影响脉络膜增殖的因素之一。②脉络膜撕脱伤后bFGF与PDGF-A在脉络膜、视网膜及玻璃体内表达高峰均出现在7~14天,此时期处于伤后细胞增殖期。③本课题为临床中脉络膜撕脱伤后玻璃体视网膜手术时机的选择、治疗干预提供参考价值。
[Abstract]:Objective: to establish a rabbit model of choroidal avulsion and observe the expression of basic fibroblast growth factor (basic fibroblast growth factor,bFGF) and platelet-derived growth factor (platelet derived growth factor-A,PDGF-A) in choroid at different time points. To explore the role of bFGF and PDGF-A in the repair of choroid proliferation after choroidal avulsion injury, and to provide a new theoretical basis for clinical treatment. Methods: 40 adult white rabbits were randomly divided into 5 groups, including normal group, 3 days, 7 days, 14 days, 28 days after injury, 8 rabbits in each group, and the experimental eyes were determined by odd and even method. A rabbit model of choroidal avulsion was established, fundus changes were observed at different time points after injury and ophthalmic B-ultrasound examination was performed, choroidal and retinal morphological changes were observed by HE staining under light microscope, and retinal morphological changes were observed at different time points after injury. Immunohistochemical SP method and enzyme-linked immunosorbent assay (Enzyme-Linked Immunosorbnent Assay,ELISA) were used to detect the expression of bFGF and PDGF-A protein in choroid, retina and vitreous. Results: the expression of bFGF and PDGF-A was up-regulated in choroid, retina and vitreous after injury, and the expression of bFGF and PDGF-A protein was up-regulated in choroid. The mean optical density of retina and the OD value of vitreous expression increased first and then decreased at the 3rd day after injury, reached the peak at the 7th day, decreased slightly at the 14th day, still maintained the high level, and decreased significantly on the 28th day after the injury, and the optical density of the retina and the OD value expressed in the vitreous were increased at first and then decreased after the injury. But higher than normal. Normal group, each experimental group, the difference was statistically significant (P0.05); 7 days and 14 days after injury, the difference was not statistically significant (P0.05), 7 days after injury and 3 days, 28 days after injury, the difference was statistically significant (P0.05). Conclusion: 1bFGF and PDGF-A are involved in the occurrence and development of choroidal proliferation and repair after choroidal avulsion injury, which is one of the factors affecting choroidal proliferation. BFGF and PDGF-A are involved in choroidal repair after choroidal avulsion injury, and bFGF and PDGF-A are involved in choroidal repair after choroidal avulsion injury. The peak of expression in retina and vitreous appeared at 7 days and 14 days, which was in the phase of cell proliferation after injury. 3 this study provided reference value for the choice of time for vitreoretinal surgery and treatment intervention after choroidal avulsion injury in clinic.
