β1整合素与声门型喉癌的关系

【摘要】: 【中文摘要】目的:检测β1整合素(intigrinβ1)在声门型喉癌和正常声带组织中的表达并进行定量分析,探讨β1整合素在声门型喉癌发生、发展中的作用,以期进一步明确声门型喉癌的发病机制,为临床防治声门型喉癌提供理论依据。材料与方法:选取经病理诊断确诊,且排除其他声带病变的声门型喉癌患者标本30例为实验组,再按分化程度分为两组,分别为高分化鳞癌组21例,中、低分化鳞癌组9例;按是否伴颈部淋巴结转移分为两组,分别为颈部淋巴结转移组8例,无颈部淋巴结转移组22例。取正常声带组织标本10例为对照组。所有标本经HE染色后,进行MaxVisionTM免疫组织化学染色,检测各组中β1整合素的表达情况,并行计算机图像定量分析。结果:1.在正常声带组织中,β1整合素的表达较弱,而在声门型喉癌组织中肿瘤细胞的表达则较强。声门型喉癌组β1整合素的阳性表达率(80.0%)高于正常声带组(60.0%),经行×列表样本构成比x2检验分析,x2=6.494,差异有统计学意义(P0.05)。2.声门型喉癌组β1整合素的阳性单位均值(33.9288±8.77570)高于正常声带组(23.8556±5.17346),差异有统计学意义(P0.05)。3.中、低分化鳞癌组β1整合素的阳性单位均值(34.9612±6.95076)虽高于高分化组(32.5840±9.49454),但差异无统计学意义(P0.05)。4.颈部淋巴结转移组β1整合素的阳性单位均值(41.1137±5.67882)高于无颈部淋巴结转移组(30.3287±7.86905),差异有统计学意义(P0.05)。结论:1.β1整合素对肿瘤细胞具有很强的特异性和敏感性,可以广泛用于肿瘤的研究。2.β1整合素在声门型喉癌组织中存在着较高的表达,与正常声带组织相比有明显差异,且差异有统计学意义。说明β1整合素表达水平增高与声门型喉癌的发生有关,β1整合素的表达与声门型喉癌呈正相关。3.β1整合素在中、低分化鳞癌组中的表达虽高于高分化鳞癌组,但差异无统计学意义,可能与样本量较小有关。β1整合素在颈部淋巴结转移组中的表达明显高于无颈部淋巴结转移组,且差异有统计学意义。表明β1整合素与声门型喉癌的淋巴结转移密切相关,提示β1整合素的表达在声门型喉癌的发生发展中起重要作用,促进了肿瘤的侵袭转移。β1整合素的表达增强可能预示着声门型喉癌的高侵袭倾向,即具有高黏附性、浸润生长、易发生转移等恶性特征,通过对β1整合素的检测,有助于评估声门型喉癌的发生、发展过程及转移倾向,可作为患者预后评估的重要指标。
[Abstract]:Objective: to detect the expression and quantitative analysis of 尾 1 integrin (intigrin 尾 1) in glottic carcinoma and normal vocal cord tissues, and to explore the role of 尾 1 integrin in the genesis and development of glottic carcinoma. In order to further clarify the pathogenesis of glottic carcinoma, to provide a theoretical basis for the clinical prevention and treatment of glottic carcinoma. Materials and methods: 30 cases of glottic laryngeal carcinoma diagnosed by pathology and excluding other vocal cord lesions were selected as experimental group and divided into two groups according to the degree of differentiation: 21 cases of well differentiated squamous cell carcinoma group, 9 cases of middle and low differentiated squamous cell carcinoma group. Patients with cervical lymph node metastasis were divided into two groups: cervical lymph node metastasis group (n = 8) and non-cervical lymph node metastasis group (n = 22). 10 normal vocal cord tissues were taken as control group. All samples were stained with HE, MaxVisionTM immunohistochemical staining was performed to detect the expression of 尾 1 integrin in each group, and the quantitative analysis of image was performed. Results: 1. The expression of 尾 1 integrin was weak in normal vocal cord tissues, but strongly expressed in glottic laryngeal carcinoma tissues. The positive expression rate of 尾 1 integrin in glottic carcinoma group (80.0%) was higher than that in normal vocal cord group (60.0%). The X-list sample composition ratio x2 test showed that the positive expression rate of 尾 1 integrin was significantly higher in glottic carcinoma group (80.0%) than that in normal vocal cord group (60.0%) (P0.05). The mean positive unit of 尾 1 integrin in glottic carcinoma group (33.9288 卤8.77570) was higher than that in normal vocal cord group (23.8556 卤5.17346), the difference was statistically significant (P0.05). The mean positive unit of 尾 1 integrin in poorly differentiated squamous cell carcinoma group (34.9612 卤6.95076) was higher than that in well differentiated group (32.5840 卤9.49454), but there was no significant difference (P0.05). The mean value of 尾 1 integrin positive unit in cervical lymph node metastasis group (41.1137 卤5.67882) was higher than that in non cervical lymph node metastasis group (30.3287 卤7.86905) (P0.05). Conclusion: 1. 尾 1 integrin has strong specificity and sensitivity to tumor cells and can be widely used in tumor studies. 2. 尾 1 integrin is highly expressed in glottic laryngeal carcinoma tissues, and 尾 1 integrin is highly expressed in glottic laryngeal carcinoma, and 尾 1 integrin is highly expressed in glottic laryngeal carcinoma. Compared with normal vocal cord tissue, there was significant difference between the two groups. The expression of 尾 1 integrin was positively correlated with glottic carcinoma. 3. The expression of 尾 1 integrin in moderate and poorly differentiated squamous cell carcinoma group was higher than that in well differentiated squamous cell carcinoma group, and the expression of 尾 1 integrin was positively correlated with glottic laryngeal carcinoma, while the expression of 尾 1 integrin in poorly differentiated squamous cell carcinoma group was higher than that in well differentiated squamous cell carcinoma group. The expression of 尾 1 integrin in cervical lymph node metastasis group was significantly higher than that in non cervical lymph node metastasis group, and the difference was statistically significant. The results suggest that 尾 1 integrin is closely related to lymph node metastasis in glottic carcinoma, suggesting that the expression of 尾 1 integrin plays an important role in the genesis and development of glottic carcinoma. The enhanced expression of 尾 1 integrin may predict the high invasion tendency of glottic carcinoma, that is, high adhesion, invasion and growth, easy metastasis and other malignant features, through the detection of 尾 1 integrin, the expression of 尾 1 integrin may indicate the high invasion tendency of glottic carcinoma, and the expression of 尾 1 integrin may indicate the high invasion tendency of glottic carcinoma. It is helpful to evaluate the occurrence, development process and metastasis tendency of glottic carcinoma, and can be used as an important index for prognosis evaluation of patients with glottic carcinoma.
