【摘要】:目的:本文旨在研究地塞米松导致的大鼠肝脏损伤中肝组织miRNA表达量的变化。方法:将Wistar大鼠随机分为两组:空白对照组(CON组,n=8),皮下注射生理盐水0.2 ml;地塞米松诱导肝损伤模型组(DEX组,n=8),皮下注射地塞米松磷酸钠注射液,2.4 mg/kg。连续注射7天后,禁食不禁水24h,处死大鼠,测定血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、血糖(GLU)及甘油三酯(TG)水平,测定肝脏组织超氧化物歧化酶(SOD)活力、丙二醛(MDA)含量和谷胱甘肽(GSH)含量。通过HE染色观察肝脏病理学变化。结果:(1)DEX组大鼠血清的ALT、AST、TG水平均显著增高,GLU水平显著降低;(2)组织切片显示,与对照组(CON)大鼠相比,注射地塞米松后大鼠肝脏细胞发生了变性;(3)通过miRNA测序分析发现,rno-miR-21-5p、rno-miR-215、 rno-miR-29b-5p、rno-miR-499-5p、rno-miR-34a-5p、rno-miR-203b-5p、 rno-miR-100-3p、rno-miR-676、rno-miR-378a-3p和一个未知的miRNA表达量显著增高,rno-miR-672-3p、rno-miR-146a-3p、rno-miR-122-5p、rno-miR-205、 mo-let-7b-5p 和 rno-miR-146b-3p表达量显著降低;poly(A) RT-qPCR检测验证了rno-miR-205、rno-let-7b-5p、rno-miR-672-3p表达量降低和rno-miR-29b-5p、 rno-miR-34a-5p、rno-miR-676表达量增高的趋势;(4)通过对差异表达miRNA进行靶基因预测和功能注释,推测到差异表达miRNA相关的生物学功能。结论:注射地塞米松造成了大鼠肝脏细胞发生变性,大鼠肝组织miRNA表达发生显著变化,差异表达的miRNA与不同的生物学功能相关。
[Abstract]:Objective: the aim of this study was to investigate the changes in the expression of miRNA in the liver tissue of rats induced by dexamethasone. Methods: Wistar rats were randomly divided into two groups: blank control group (group CON, n=8), subcutaneous injection of normal saline 0.2 ml, dexamethasone induced liver injury model group (DEX group, n=8), subcutaneous injection of Dexamethasone Sodium Phosphate Injection, 2.4 mg/kg. After 7 days of continuous injection, fasting 24h, rats were killed, serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), blood glucose (GLU) and triglyceride (TG) level, the determination of liver tissue superoxide dismutase (SOD) activity, malondialdehyde (MDA) content and glutathione (GSH) content. The pathological changes of liver were observed by HE staining. Results: (1) serum DEX group rats ALT, AST and TG levels were significantly increased, GLU levels decreased significantly; (2) tissue sections showed that, compared with the control group (CON) compared with rats after injection of dexamethasone in rats liver cell degeneration; (3) by miRNA sequencing analysis in this paper. Rno-miR-21-5p, rno-miR-215, rno-miR-29b-5p, rno-miR-499-5p, rno-miR-34a-5p, rno-miR-203b-5p, rno-miR-100-3p, rno-miR-676, rno-miR-378a-3p and an unknown miRNA expression was significantly increased, the expression of rno-miR-672-3p, rno-miR-146a-3p, rno-miR-122-5p, rno-miR-205, mo-let-7b-5p and rno-miR-146b-3p were significantly reduced; poly (A) RT-qPCR, rno-let-7b-5p, rno-miR-205 detection verified the expression of rno-miR-672-3p decreased and rno-miR-29b-5p, rno-miR-34a-5p and the expression of rno-miR-676 increased; (4) the difference in expression of miRNA Target gene prediction and functional annotation were used to speculate on the biological functions of differential expression of miRNA. Conclusion: the injection of dexamethasone induces the degeneration of rat liver cells, and the expression of miRNA is significantly changed in rat liver. The differential expression of miRNA is related to different biological functions.
