甲磺酸多黏菌素对大鼠巨噬细胞免疫功能的影响

【摘要】:目的探究甲磺酸多黏菌素E(CMS)对大鼠腹腔巨噬细胞免疫功能的影响。方法用噻唑蓝(MTT)法检测大鼠腹腔巨噬细胞在细胞悬液(阴性对照组)和25,50,100,125,250,500μg·m L~(-1)CMS作用24 h后的细胞存活率。用酶联免疫法测定50,125μg·m L~(-1)CMS作用3,6,12,24 h后,巨噬细胞肿瘤坏死因子-α(TNF-α)的分泌水平。用免疫荧光微球法测定50,125μg·m L~(-1)CMS作用3,6,12 h后,巨噬细胞的吞噬功能。结果与阴性对照组100.00%相比,25,50,100,250,500μg·m L~(-1)CMS作用后的细胞存活率分别为118.46%,124.72%,122.54%,46.64%,13.06%,差异均有统计学意义(P0.01);125μg·m L~(-1)CMS作用后的细胞存活率为102.60%,差异无统计学意义(P0.05)。作用3 h后,阴性对照组与125μg·m L~(-1)CMS给药组的TNF-α水平分别为32.51 pg·m L~(-1),56.09 pg·m L~(-1),差异有统计学意义(P0.05)。作用6 h后,阴性对照组与50,125μg·m L~(-1)CMS给药组的吞噬百分率分别为70.93%,82.03%,80.80%,阴性对照组与50,125μg·m L~(-1)CMS给药组比较差异均有统计学意义(P0.01)。结论 CMS能通过提高细胞活性、促进细胞因子分泌及增强细胞吞噬功能等途径增强巨噬细胞的免疫功能,进而发挥其免疫调节作用。
[Abstract]:Objective to investigate the effect of polymyxine mesylate (PMS) on the immune function of peritoneal macrophages in rats. And 25. The cell survival rate of 50,100,125,250,500 渭 g 路mL ~ (-1) CMS for 24 h was determined by enzyme-linked immunosorbent assay (Elisa). The levels of tumor necrosis factor- 伪 (TNF- 伪) in macrophages were determined by immunofluorescence microsphere method. The phagocytic function of macrophages was observed after 12 h treatment with 125 渭 g 路m ~ (-1) CMS. Results compared with the negative control group (100.00%), the phagocytic function of macrophages was 2550 / 100. The cell survival rate of 250,500 渭 g 路m ~ (-1) ~ (-1) CMS was 118.46% and 124.72% and 122.54 ~ 46.64%, respectively. 13.06, the difference was statistically significant (P 0.01); The cell survival rate was 102.60 after treatment with 125 渭 g 路mL ~ (-1) CMS, but there was no significant difference between the two groups (P 0.05). After 3 h of treatment, the cell survival rate was 102.60 渭 g 路mL ~ (-1). The levels of TNF- 伪 in the negative control group and 125 渭 g 路mL ~ (-1) CMS group were 32.51 PG 路m ~ (-1). 56.09 PG 路mL ~ (-1), the difference was statistically significant (P 0.05). After 6 hours of treatment, the negative control group was 50%. The percentage of phagocytosis in 125 渭 g 路mL ~ (-1) CMS group was 70.93 and 82.03, respectively, and that in negative control group was 80.80, and that in negative control group was 50. 125 渭 g 路mL ~ (-1) CMS could improve cell activity by increasing the cell activity, and the difference was statistically significant (P 0.01). Promoting cytokine secretion and enhancing phagocytic function can enhance the immune function of macrophages and then exert its immunomodulatory effect.
