微波消解-电感耦合等离子体质谱法测定大鼠全血中纳米银

【摘要】:目的:建立和确证一种简便、准确的测定大鼠全血中银浓度的分析方法。方法:选取铟(115In)为内标,先用硝酸对全血样品进行微波消解处理,再采用电感耦合等离子体质谱法测定消化液中银离子的浓度。结果:内源性物质不干扰银和内标的测定,全血中银的浓度在5.0~1 000μg·L-1范围内线性关系良好,日内精密度RSD≤3.3%,日间精密度RSD≤3.8%;提取回收率为97.5%~101.9%,基质效应为97.4%~99.4%。应用该法研究了大鼠静脉注射纳米银后银的体内经时变化,结果表明,给药(40 nm)后,大鼠血液中银浓度先迅速下降而后上升,之后再缓慢消除。结论:该方法可为纳米银在生物机体内的评价研究提供分析方法参考。
[Abstract]:Objective: to establish and confirm a simple and accurate method for the determination of silver in whole blood of rats. Inductively coupled plasma mass spectrometry (ICP-MS) was used to determine the concentration of silver ion in digestive fluid.
