右美托咪定对离体大鼠肠系膜动脉炎性相关蛋白表达的影响

【摘要】:目的研究右美托咪定(DEX)对大鼠肠系膜动脉肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)及凋亡相关因子Caspase-3表达e笞饔谩7椒ㄈPF级SD雄性大鼠,脱臼处死后,在体视显微镜下分离取出肠系膜动脉。建立脂多糖(LPS)诱导的血管损伤实验模型,并根据随机化原则将损伤血管分为DEX组和对照组。DEX组根据所给予DEX的浓度不同分为10~(-8)、10~(-7)、10~(-6) mol/L 3个亚组。分离提取各组动脉组织中RNA及总蛋白后,RT-PCR方法检测TNF-α、IL-1β及CaSR等因子的mRNA水平;Western blot检测TNF-α、CaSR、AMPK及Caspase-3等蛋白的表达变化。结果 10~(-8)、10~(-7)、10~(-6) mol/L DEX组中LPS诱导的血管炎性反应均明显降低,TNF-α的mRNA和蛋白表达及IL-1β的mRNA表达减弱。DEX组血管条的Caspase-3蛋白表达较LPS组显著降低。DEX对LPS诱导的血管AMPK蛋白表达和CaSR的mRNA表达没有明显作用。结论 DEX明显降低与炎症相关蛋白的表达,提示DEX有一定的抗炎症作用。
[Abstract]:Objective to study the effect of dexmetomidine (DEX) on the expression of tumor necrosis factor- 伪 (TNF- 伪) and apoptosis related factor (Caspase-3) in mesenteric artery of rats. Male SD rats of grade PF were killed by dislocations. Mesenteric arteries were isolated and removed under stereoscopic microscope. An experimental model of vascular injury induced by lipopolysaccharide (LPS) was established. According to the principle of randomization, the injured vessels were divided into three groups: DEX group and control group. DEX group was divided into three subgroups according to the concentration of DEX. RNA and total protein were isolated and extracted from the arterial tissues of each group. RT-PCR was used to detect TNF- 伪 IL-1 尾 and CaSR. The expression of TNF- 伪 -CaSR-AMPK and Caspase-3 were detected by Western blot. Results in the mol/L DEX group, the expression of mRNA and protein and the mRNA expression of IL-1 尾 were significantly decreased in the vascular strips of the mol/L DEX group. Compared with LPS group, DEX had no significant effect on the expression of AMPK protein and mRNA expression of CaSR induced by LPS. Conclusion DEX can significantly decrease the expression of inflammatory related protein. The results suggest that DEX has some anti-inflammatory effect.
