不同血样处理方式对大鼠部分血生化指标的影响

【摘要】：目的:观察不同血样处理方式对大鼠生化指标的影响,为临床前动物试验中应用血浆或稀释血清进行生化指标的测定提供参考。方法:取20只空白SD大鼠,采血并分离得到EDTA-K2抗凝血浆、枸橼酸钠抗凝血浆、非抗凝血清和稀释血清(用去离子水1∶1稀释),采用全自动生化分析仪进行13项生化指标的测定,检测结果采用配对T检验进行统计学比较和分析。结果:与血清检测值相比,EDTA抗凝血浆的AST,ALP,LDH和CK均明显降低(P0.05);枸橼酸钠抗凝血浆的13项检测值均明显降低,在乘以系数1.1后也存在AST,TP,LDH和CK明显降低(P0.05);与血清原液检测值相比,稀释后血清的Alb升高(P0.05),TBil降低(P0.05)。结论:药物临床前研究中推荐采用血清进行生化指标测定,在必需采用抗凝血浆进行生化指标测定时,建议不要采用枸橼酸钠抗凝血浆,可采用EDTA-K2抗凝血浆,同时要注意到AST,ALP,LDH和CK等指标的血浆结果与血清检测值缺乏可比性。
[Abstract]:Objective: to observe the effect of different blood samples on the biochemical indexes of rats, and to provide reference for the determination of biochemical indexes by using plasma or diluted serum in preclinical animal test.Methods: EDTA-K2 anticoagulant plasma and sodium citrate anticoagulant plasma were obtained from 20 SD rats.Non-anticoagulant serum and diluted serum (diluted with deionized water at 1:1) 13 biochemical indexes were measured by automatic biochemical analyzer. The results were compared and analyzed statistically by paired T test.Results: compared with the serum detection value, the LDH and CK of the anticoagulant plasma of EDTA decreased significantly (P 0.05), the 13 items of the anticoagulant plasma of sodium citrate decreased obviously, and the LDH and CK of the anticoagulant plasma were significantly decreased after multiplying by the coefficient 1.1.The level of Alb in serum was increased after dilution, and the level of TBil in serum was lower than that in control group (P 0. 05 and P 0. 05).Conclusion: it is recommended that serum be used to determine biochemical indexes in preclinical study. When anticoagulant plasma is necessary for biochemical index determination, sodium citrate anticoagulant plasma should be used instead of EDTA-K2 anticoagulant plasma.At the same time, we should note that the results of plasma LDH and CK were not comparable with those of serum.
