氯吡格雷对血小板聚集及肝细胞色素P4503A4表达的影响

【摘要】：目的探讨氯吡格雷对大鼠血小板聚集及肝细胞色素P4503A4表达的影响。方法 45只Wistar大鼠按照体重随机分为3组(空白组,对照组和实验组),每组15只。空白组大鼠灌胃生理盐水5 m L·kg-1·d-1,对照组灌胃阿司匹林100 mg·kg-1·d-1,实验组大鼠灌胃氯吡格雷20 mg·kg-1·d-1,均5m L,3组大鼠均处理7 d。药物处理7 d后,心脏取血,检测血小板聚集率和血小板聚集抑制率。Western blot法检测3组肝细胞色素P4503A4表达水平。结果实验组大鼠血小板最大聚集率为(37.23±5.01)%,显著低于空白组的(57.21±5.23)%和对照组的(47.23±4.21)%(P0.05);实验组大鼠血小板聚集抑制率为(33.56±4.64)%,显著高于对照组的(21.12±3.36)%(P0.05);与空白组和对照组相比,实验组大鼠肝细胞色素P4503A4表达水平显著降低(P0.05)。结论氯吡格雷可能通过抑制大鼠肝细胞色素P4503A4表达来达到抗血小板功能的作用。
[Abstract]:Objective to investigate the effects of clopidogrel on platelet aggregation and hepatic cytochrome P4503A4 expression in rats.Methods 45 Wistar rats were randomly divided into 3 groups according to their body weight: blank group, control group and experimental group with 15 rats in each group.Rats in the blank group were given normal saline 5 mL kg-1 d-1, the control group was given aspirin 100mg kg-1 d-1, the experimental group was treated with clopidogrel 20 mg kg-1 d-1, and the rats in the control group were treated with clopidogrel 20 mg kg-1 d-1 for 7 days.After 7 days of treatment, blood was taken from the heart, platelet aggregation rate and platelet aggregation inhibition rate.
