表面活性剂对活跃链霉菌产那西肽发酵的影响

【摘要】:目的研究Triton X-100、Ca Cl2、DMSO、山梨醇、SDS、Tween-80、CTAB等表面活性剂对活跃链霉菌发酵产那西肽的影响。方法采用单因素实验研究活跃链霉菌的基础代谢以及表面活性剂加入时间对那西肽效价的影响;采用响应面法对表面活性剂添加浓度进行优化。结果基础代谢研究表明,活跃链霉菌产那西肽合成相对滞后于菌体生长。多种表面活性剂均显著影响那西肽的生物合成,整体表现为低浓度促进、高浓度抑制。表面活性剂依其类型及加入时机的不同,对那西肽产量的影响表现出组间类型差异性及组内时间依赖性。结论 DMSO、Ca Cl2、Triton X-100在低浓度范围内对那西肽产量的促进效果最为显著;分别在24、24和48h以DMSO 0.808mg/m L、Ca Cl2 0.799mg/m L和Triton X-100 0.574mg/m L的浓度为最佳添加浓度,该条件下那西肽效价达到931.493μg/m L,比优化前提高了7.73%。
[Abstract]:Methods the basic metabolism of Streptomyces activis and the effect of surfactant addition time on the titer of natritin were studied by single factor experiment, and the concentration of surfactant was optimized by response surface method.Results the basic metabolism study showed that the synthesis of nacetin produced by Streptomyces activis lagged behind the growth of mycelium.Many surfactants significantly affected the biosynthesis of natritin, which was promoted by low concentration and inhibited by high concentration.The effect of surfactants on the yield of natritin was different among groups and time-dependent according to the type and timing of addition of surfactants.Conclusion DMSO 0.808mg/m Ca Cl2 0.799mg/m L and Triton X-100 0.574mg/m L were the most effective in promoting the yield of natritin in the low concentration range, and the optimal concentrations were DMSO 0.808mg/m Ca Cl2 0.799mg/m L and Triton X-100 0.574mg/m L at 24 h and 48 h, respectively.The titer of nazeptide reached 931.493 渭 g / mL, which was 7.73% higher than that before optimization.
