建立测定兔眼角膜中盐酸特比萘芬含量的甲醇浸提-HPLC法

【摘要】:目的:建立测定兔眼角膜中盐酸特比萘芬含量的方法。方法:取日本大耳白兔18只,随机分为6组,每组3只,双眼给予0.2%盐酸特比萘芬滴眼液50μl。分别于给药后15、30、60、120、240、480 min时处死兔,剖取角膜,加入甲醇,充氮气,密封冷藏静置浸泡24 h后,涡旋,离心,取上清液采用高效液相色谱法测定其中盐酸特比萘芬的含量。色谱柱为Waters X-Bridge C18,流动相为甲醇-水(92∶8),流速为0.7 ml/min,检测波长为224 nm,柱温为40℃,进样量为20μl。结果:在兔眼角膜醇浸提液中盐酸特比萘芬检测质量浓度在0.007 64~0.382μg/ml(r=0.999 8,n=6)范围内与峰面积呈良好的线性关系;方法回收率为99.6%(RSD=4.5%,n=3),提取回收率为98.5%~103.2%(RSD=6.8%~11.0%,n=3);日内RSD为4.1%(n=5),日间RSD为4.5%(n=5)。给药480 min后仍可测得盐酸特比萘芬,其质量浓度为(0.102±0.043 3)μg/ml。结论:本方法专属性强、准确度高,可用于兔眼角膜中盐酸特比萘芬的含量测定。
[Abstract]:Objective: to establish a method for the determination of terbinafine hydrochloride in rabbit cornea.Methods: eighteen Japanese white rabbits were randomly divided into 6 groups, 3 rabbits in each group, and the eyes were given 0.2% terbinafine hydrochloride eye drops (50 渭 l).Rabbits were killed at 15: 30 and 60120 240 480 min after administration, cornea were cut off, methanol was added, nitrogen was filled, immersion was kept in cold storage for 24 h, vortex, centrifugation, supernatant was taken and the content of terbinafine hydrochloride was determined by high performance liquid chromatography (HPLC).The chromatographic column was Waters X-Bridge C18, the mobile phase was methanol-water (92: 8), the flow rate was 0.7 ml / min, the detection wavelength was 224nm, the column temperature was 40 鈩,
