建立测定兔眼角膜中盐酸特比萘芬含量的甲醇浸提-HPLC法

【摘要】：目的:建立测定兔眼角膜中盐酸特比萘芬含量的方法。方法:取日本大耳白兔18只,随机分为6组,每组3只,双眼给予0.2%盐酸特比萘芬滴眼液50μl。分别于给药后15、30、60、120、240、480 min时处死兔,剖取角膜,加入甲醇,充氮气,密封冷藏静置浸泡24 h后,涡旋,离心,取上清液采用高效液相色谱法测定其中盐酸特比萘芬的含量。色谱柱为Waters X-Bridge C18,流动相为甲醇-水(92∶8),流速为0.7 ml/min,检测波长为224 nm,柱温为40℃,进样量为20μl。结果:在兔眼角膜醇浸提液中盐酸特比萘芬检测质量浓度在0.007 64~0.382μg/ml(r=0.999 8,n=6)范围内与峰面积呈良好的线性关系;方法回收率为99.6%(RSD=4.5%,n=3),提取回收率为98.5%~103.2%(RSD=6.8%~11.0%,n=3);日内RSD为4.1%(n=5),日间RSD为4.5%(n=5)。给药480 min后仍可测得盐酸特比萘芬,其质量浓度为(0.102±0.043 3)μg/ml。结论:本方法专属性强、准确度高,可用于兔眼角膜中盐酸特比萘芬的含量测定。
