高效毛细管电泳法测定大鼠血浆中头孢地尼浓度及其药动学研究

【摘要】:目的:测定大鼠血浆中头孢地尼浓度,并进行药动学考察。方法:采用高效毛细管电泳法(HPCE)。使用熔融硅胶毛细管柱(75μm,总长为30 cm,有效长度为21.5 cm),缓冲液为20 mmol/L柠檬酸,进样电压为10 k V持续10 s,分离电压为-20 k V,检测波长为214 nm。取6只大鼠ig头孢地尼溶液(20 mg/kg),分别于给药前及给药后0.25、0.5、1、1.5、2、3、4、6、12、24 h经尾缘静脉取血0.2 m L进行测定。采用BAPP 2.0软件计算药动学参数。结果:头孢地尼质量浓度在0.2~50μg/m L范围内线性关系良好(r=0.999 7),定量下限为0.2μg/m L;日内(n=6)、日间(n=3)精密度试验的RSD均不大于12.2%;稳定性试验的RSD≤9.10%(n=6);方法回收率为95.4%~114.3%(RSD=9.0%,n=6);基质效应为63.5%~70.2%(RSD=10.39%,n=6)。头孢地尼在大鼠体内的t1/2为(0.54±0.01)h,MRT为(1.90±0.14)h,c_(max)为(32.92±0.81)μg/m L,t_(max)为(1.50±0.02)h,AUC_(0-24 h)为(46.65±0.44)μg·h/m L,AUC_(0-∞)为(46.83±0.44)μg·h/m L。结论:该方法快速、准确、简便,可用于大鼠血浆中头孢地尼浓度的测定及其药动学考察。
[Abstract]:Objective: to determine the concentration of cefdinir in rat plasma and to investigate its pharmacokinetics.Methods: high performance capillary electrophoresis (HPCE) was used.The fused silica gel capillary column was used for 75 渭 m, the total length was 30 cm, the effective length was 21.5 cm ~ (-1), the buffer solution was 20 mmol/L citric acid, the injection voltage was 10 kV for 10 s, the separation voltage was -20 kV, the detection wavelength was 214 nm.Before and after administration of cefdinir solution, 0.2 mL of blood was collected from 6 rats before and after administration of cefodienil (0.25g / kg), respectively.
