一株银杏内生真菌抗菌代谢产物的分离与鉴定

【摘要】:目的分离并鉴定银杏内生真菌产生的抗菌活性物质。方法在一株镰刀菌属内生真菌的发酵液中检测出了较强的抗革兰阳性菌的活性,抗菌活性筛选的检定菌为短小芽孢杆菌[CMCC(B)63202]。发酵液经液液萃取、硅胶柱层析、高效液相分离得到化合物A和B。液相色谱条件:以Shim-pack C_(18)(250mm×10mm,5μm)为固定相,以甲醇-水(5:6)为流动相,流速1.0m L/min,检测波长254nm,进样量20μL。结果经结构解析,化合物A的分子量为639,由3个α-羟基-β-甲基-丁酸(Hiv)与3个N甲基缬氨酸(MeVal)相互间隔地缩合成一个环状对称结构,与文献中恩镰孢菌素B的结构一致。化合物B的分子量为653,与恩镰孢菌素B相差一个CH_2,推测与恩镰孢菌素B_1或恩镰孢菌素D同质。结论该银杏内生真菌产生的抗菌活性物质为恩镰孢菌素。
[Abstract]:Objective to isolate and identify the antimicrobial active substances produced by endophytic fungi of Ginkgo biloba. Methods A strain of Fusarium endophytic fungi was used to detect the activity of Gram-positive bacteria in the fermentation broth of Fusarium. The antimicrobial activity was screened by Bacillus pumilus [CMCC(B)63202]. Compounds A and B were separated from fermentation broth by liquid-liquid extraction, silica gel column chromatography and high performance liquid chromatography. The liquid chromatographic conditions were as follows: Shim-pack C_(18)(250mm 脳 10mm-1 5 渭 m) as stationary phase, methanol-water 5: 6) as mobile phase, flow rate 1.0 mL / min, detection wavelength 254 nm, sample volume 20 渭 L. Results the molecular weight of compound A was 639. Three 伪 -hydroxy- 尾 -methyl-butyric acid (Hiv) and three N-methyl valine (MeVala) were separated to form a cyclic symmetric structure, which was consistent with the structure of Fusarium B in the literature. The molecular weight of compound B is 653, which is one CHS _ 2 difference from that of Fusarium B, which is conjectured to be the same as Fusarium B _ 1 or E _ S _ D. Conclusion the antimicrobial activity produced by endophytic fungi of Ginkgo biloba is Fusarium mongolicum.
