桦褐孔菌多糖IOP3a乙酰化修饰及其抗氧化活性

【摘要】:以桦褐孔菌多糖IOP3a为原料,采用乙酸酐法制备乙酰化桦褐孔菌多糖(Ac-IOP3a),以乙酰化取代度为指标,考察乙酸酐用量、反应温度和反应时间对取代度的影响。在单因素试验的基础上,采用响应面法对IOP3a乙酰化工艺进行优化,并通过Ac-IOP3a对·OH、DPPH·和O_2~-·的清除作用探讨其抗氧化活性。结果表明,桦褐孔菌多糖IOP3a乙酰化最佳工艺为:乙酸酐用量3.3 mL(固定IOP3a多糖量100 mg),反应时间3.1 h,反应温度64℃,在此条件下,Ac-IOP3a的取代度为0.416。抗氧化研究表明:与IOP3a多糖相比,Ac-IOP3a多糖的抗氧化活性明显增强。
[Abstract]:Ac-IOP3a was prepared by acetic anhydride method from IOP3a. The effects of acetic anhydride dosage, reaction temperature and reaction time on the degree of substitution were investigated. On the basis of the single factor experiment, the IOP3a acetylation process was optimized by response surface method, and the antioxidant activity of Ac-IOP3a was investigated by the scavenging effect of Ac-IOP3a on OH-DPPH and O _ 2P _ 2 ~ (-). The results showed that the optimum conditions of IOP3a acetylation were as follows: the amount of acetic anhydride was 3.3 mL, the reaction time was 3.1 h, the reaction temperature was 64 鈩,
