透明质酸修饰的尿酸酶多囊脂质体体外稳定性初步研究

【摘要】:目的:制备了透明质酸修饰的尿酸酶多囊脂质体(hyaluronic acid-uricase liposomes,UHMVL),并且对尿酸酶(uricase,UC)以及UHMVL在体外的活性和稳定性进行了初步的研究。方法:采用复乳法制备了UHMVL,并且测定了其包封率。然后从最适温度(n=3)、最适p H(n=3)、体外稳定性(n=3)初步考察了UHMVL和UC的差异。结果:UHMVL的包封率为(62.48±3.87)%。UHMVL及UC最适温度均为40℃;UHMVL的最适p H值为8.0,UC的最适p H值为8.5。稳定性测定结果表明,UHMVL体外稳定性均比UC好。UHMVL在提高了UC在体外活性的同时,也明显地增强了UC在体外的稳定性。结论:UHMVL在提高了UC体外活性的同时,也明显地增强了游离酶UC在体外的稳定性。
[Abstract]:Aim: to prepare hyaluronic acid-uricase liposome UHMVLA modified by hyaluronidase, and to study the activity and stability of uricase (UC) and UHMVL in vitro. Methods: UHMVLs were prepared by double emulsion method and their encapsulation efficiency was determined. Then, the difference between UHMVL and UC was preliminarily investigated from the optimum temperature, the optimum temperature, the optimum pH, the in vitro stability and the in vitro stability.
