微波加热顺铂还原铁粉混悬液对猪肝细胞凋亡的影响

【摘要】:【目的】 探讨微波加热顺铂还原铁粉混悬液诱导猪肝细胞凋亡及其与TNF-α、Bcl-2和Bax蛋白表达的关系。 【方法】 将猪随机地分成对照组和微波治疗组。微波治疗组用穿刺针将顺铂还原铁粉混悬液注入消融治疗后的猪肝形成的空洞内,用医用微波治疗仪局部加热,对照组则不予处理。采用原位末端转移酶标记(TUNEL)技术分析检测细胞的凋亡情况,免疫组化法检测细胞凋亡相关基因TNF-α、Bcl-2和Bax蛋白的表达水平。 【结果】 1、微波治疗组和对照组猪肝内均可见凋亡细胞,微波治疗组中平均凋亡指数(%)为7.55±1.74,对照组中平均凋亡指数(%)为5.33±1.45;两者相比较,微波治疗组细胞平均凋亡指数(%)明显高于对照组,差异有统计学意义(P0.01)。 2、微波治疗组中TNF-α蛋白平均阳性表达率(%)为11.70±3.02,对照组中TNF-α蛋白平均阳性表达率(%)为8.20±1.43;两者相比较,微波治疗组TNF-α蛋白阳性表达率明显高于对照组,,差异有统计学意义(P0.01)。 3、微波治疗组中Bcl-2蛋白平均阳性表达率(%)为6.61±1.74,对照组中Bcl-2蛋白平均阳性表达率(%)为9.81±3.01;两者相比较,微波治疗组Bcl-2蛋白平均阳性表达率较对照组明显降低,差异有统计学意义(P0.05)。 4、微波治疗组中Bax蛋白平均阳性表达率(%)为13.37±3.40,对照组中Bax蛋白平均阳性表达率(%)为9.81±2.80,两者相比较,微波治疗组Bax蛋白平均阳性表达率较对照组明显升高,差异有统计学意义(P0.05)。 5、微波治疗组中Bax/Bcl-2平均比值为5.34±2.55,对照组中Bax/Bcl-2平均比值为3.29±1.59;两者相比较,微波治疗组Bax/Bcl-2平均比值较对照组明显升高,差异有统计学意义(P0.05)。 【结论】 1、微波加热顺铂还原铁粉混悬液能有效地诱导猪肝细胞凋亡。 2、微波加热顺铂还原铁粉混悬液能有效地诱导猪肝细胞凋亡的机制可能是通过以下几条途径来实现的: 1)增加TNF-α蛋白的表达; 2)抑制Bcl-2蛋白的表达; 3)增加Bax蛋白的表达; 4)提高Bax/Bcl-2的比值。
[Abstract]:[purpose] To investigate the relationship between the apoptosis of porcine hepatocytes induced by Cisplatin reduced iron powder suspension and the expression of TNF- 伪 Bcl-2 and Bax protein. [methods] Pigs were randomly divided into control group and microwave treatment group. In the microwave treatment group, Cisplatin reduced iron powder suspension was injected into the cavity of the pig liver after ablation with puncture needle, and the local heating was used with the medical microwave therapy instrument, while the control group was not treated. Apoptosis was detected by in situ terminal transferase labeling (Tunel) technique and the expression of apoptosis related gene TNF- 伪 and Bax protein was detected by immunohistochemistry. [results] 1. Apoptotic cells were found in the liver of pigs in microwave treatment group and control group, the average apoptosis index in microwave treatment group was 7.55 卤1.74, and the average apoptosis index in control group was 5.33 卤1.45.The mean apoptotic index in microwave treatment group was significantly higher than that in control group. The difference was statistically significant (P 0.01). (2) the average positive expression rate of TNF- 伪 protein in microwave treatment group was 11.70 卤3.02, and that in control group was 8.20 卤1.43.The positive rate of TNF- 伪 protein expression in microwave treatment group was significantly higher than that in control group (P 0.01). 3. The average positive expression rate of Bcl-2 protein was 6.61 卤1.74 in microwave treatment group and 9.81 卤3.01 in control group. Compared with control group, the average positive rate of Bcl-2 protein in microwave treatment group was significantly lower than that in control group (P 0.05). 4. The average positive expression rate of Bax protein was 13.37 卤3.40 in microwave group and 9.81 卤2.80 in control group. Compared with the control group, the average positive rate of Bax protein in microwave treatment group was significantly higher than that in control group (P 0.05). 5, the average ratio of Bax/Bcl-2 in microwave treatment group was 5.34 卤2.55, and that in control group was 3.29 卤1.59.The average ratio of Bax/Bcl-2 in microwave treatment group was significantly higher than that in control group (P 0.05). [conclusion] 1. Microwave heating of Cisplatin reduced iron powder suspension could effectively induce the apoptosis of porcine hepatocytes. 2. Microwave heating of Cisplatin reduced iron powder suspension can effectively induce the apoptosis of porcine hepatocytes. 1) increase the expression of TNF- 伪 protein; 2) inhibiting the expression of Bcl-2 protein; 3) increase the expression of Bax protein; 4) increasing the ratio of Bax/Bcl-2.
