粘质沙雷菌毒性噬菌体PS4的生物学特性及其在小鼠血清中的杀菌作用研究

【摘要】:目的研究粘质沙雷菌毒性噬菌体PS4的生物学特性及其在小鼠血清中的抗菌活性。方法观察PS4的噬菌斑及电镜下形态;测定PS4的噬菌谱、生长曲线及其理化稳定性,PS4在营养肉汤培养基、新鲜小鼠血清和灭活小鼠血清中的杀菌作用,以及小鼠血清中影响PS4杀菌能力影响因素。结果 PS4为长尾型噬菌体,在以粘质沙雷菌临床株S4为指示菌时,PS4可形成直径为1~1.5mm完全透明噬菌斑,其感染S4的潜伏期为10min,暴发量约为100PFU/cell,且S4对PS4的抗性突变率仅为10-7。PS4在pH 5~10的环境中具有较好的稳定性,在50℃的环境中仍能长时间保持活力。PS4在新鲜未灭活血清中对S4不具杀菌能力,但在56℃热灭活30min的鼠血清中以及在营养肉汤中的杀菌率均达99.99%,差异无统计学意义(P0.05)。结论 PS4具有一定的酸碱稳定性和热稳定性,有望用于医院环境"消毒",以减少多重耐药粘质沙雷菌所致的院内感染的发生。但因其杀菌作用受血清中不耐热成分的严重干扰,故不能用于治疗由S4引起的小鼠全身感染。
[Abstract]:Objective to study the biological characteristics of bacteriophage PS4 and its antibacterial activity in mouse serum. Methods the plaque of PS4 and its morphology under electron microscope were observed, the bacteriophage spectrum, growth curve and physicochemical stability of PS4 were determined, and the bactericidal effects of PS4 in nutrient broth medium, fresh mouse serum and inactivated serum were determined. And the influence factors of PS4 bactericidal ability in serum of mice. Results PS4 was a long-tailed bacteriophage. When S4, a clinical strain of Shareh, was used as indicator, PS4 could form a completely transparent plaque with diameter of 1~1.5mm. The incubation period of S4 infection was 10 min, and the number of outbreaks was about 100 PFU / p cell. The mutation rate of S4 resistance to PS4 was only stable in pH 5 ~ 10 environment.
