丙泊酚麻醉对创伤性脑损伤大鼠神经元再生和神经功能的影响

【摘要】:目的观察丙泊酚麻醉对创伤性脑损伤大鼠(TBI)的影响。方法用Feeney法建立TBI模型。按照体重将雄性Wistar大鼠分为4组:假手术组、模型组和低高2个剂量实验组,每组20只。低高2个剂量实验组尾静脉注射丙泊酚36,72 mg·kg~(-1)·h~(-1)。术后28 d,比较4组大鼠术后的死亡率,用平衡测验方法检测平衡能力,用巴恩斯迷宫法检测学习记忆能力,用免疫组化法检测神经元新生状况。结果术后28 d,低高2个剂量实验组大鼠死亡率分别为30%,40%,与模型组比较差异均有统计学意义(均P0.05)。高剂量实验组大鼠进入目的箱时间为(169.64±6.77)s,显著高于模型组的(144.21±8.52)s,差异有统计学意义(P0.05);高剂量实验组平衡能力为(37.29±4.26)s,显著低于模型组的(49.37±5.17)s,差异有统计学意义(P0.05)。模型组大鼠的神经元新生数为(680.46±65.17)个,低高2个剂量实验组大鼠分别为(364.59±76.21),(302.14±42.77)个,与模型组比较差异均有统计学意义(均P0.01)。结论丙泊酚可降低创伤性脑损伤大鼠神经元再生,且对创伤性脑损伤大鼠具有恶化作用。
[Abstract]:Objective to observe the effect of propofol anesthesia on TBI in rats with traumatic brain injury. Methods TBI model was established by Feeney method. Male Wistar rats were divided into 4 groups according to their body weight: sham operation group, model group and low and high dose groups, 20 rats in each group. Two doses of propofol were injected into caudal vein of the two experimental groups with propofol 72 mg / kg 路kg ~ (-1) 路h ~ (-1) 路L ~ (-1) 路h ~ (-1). On the 28th day after operation, the mortality rate of the four groups was compared. Balance test was used to test the balance ability, Barnes maze method was used to detect the ability of learning and memory, and immunohistochemical method was used to detect the status of neuronal regeneration. Results on the 28th day after operation, the mortality of the rats in the experimental group with low dose and high dose was 30 and 40, respectively, which was significantly different from that of the model group (all P 0.05). The time of entering the box in the high dose group was 169.64 卤6.77 s, which was significantly higher than that in the model group (144.21 卤8.52 s, P 0.05), and the balance ability of the high dose group was 37.29 卤4.26 seconds, which was significantly lower than that in the model group (49.37 卤5.17 s). The number of neurons in the model group was 680.46 卤65.17, and that in the low and high dose groups was 364.59 卤76.21, 302.14 卤42.77, respectively, which was significantly different from that in the model group (P 0.01). Conclusion propofol can decrease the neuronal regeneration in rats with traumatic brain injury and has a worsening effect on traumatic brain injury.
