咪达唑仑糖浆混合液稳定性研究

【摘要】:目的:探究我院自行配制咪达唑仑糖浆混合液在中国人群中的口感接受程度以及稳定性。方法:咪达唑仑盐酸注射液5mg/ml(M)与单糖浆850mg/l(S)按照不同体积配比混合制成5组不同浓度的咪达唑仑糖浆混合液(对照组5mg/mlM:S=1:0;A 组 3.3mg/mlM:S=1:0.5;B 组 2.5mg/mlM:S=1:1;C 组 2.0mg/mlM:S=1:1.5;D组1.67mg/mlM:S=1:2),采用滴定法将不同浓度咪达唑仑糖浆混合液按照随机顺序依次滴定于40名中国健康成人志愿者舌根部进行品尝,志愿者每品尝完一组立即漱口 60秒,并对该组咪达唑仑糖浆混合液口感选择接受程度并打分,统计出接受程度最高的一组。配置接受程度最高组咪达唑仑糖浆混合液分别储存于4℃和常温下,采用质谱液相联用法测量配置后第0,8,9,10,12,14,21天咪达唑仑糖浆混合液中咪达唑仑的药物浓度,pH值测量仪测量咪达唑仑糖浆混合液pH值,肉眼观察咪达唑仑糖浆混合液的颜色、澄明度及外观的变化。结果:1、口感:咪达唑仑注射液与单糖浆体积配比为1:2(即咪达唑仑糖浆混合液浓度1.67mg/ml)时口感接受程度最高,接受程度评分显著高于其他组,差异有统计学意义(P0.05)。2、稳定性:1)咪达唑仑糖浆混合液21天内浓度稳定性良好,常温与4℃下浓度差异无统计学意义(P0.05)。2)咪达唑仑糖浆混合液21天内pH值呈线性增大趋势,4℃相对于室温pH值更稳定,差异有统计学意义(P0.05),建议冷藏保存。3)咪达唑仑糖浆混合液21天内物理性状稳定性良好。结论:北京协和医院自行配制的咪达唑仑糖浆混合液口感舒适,稳定性良好,可作为一种良好的咪达唑仑注射液替代口服剂应用于中国临床。
[Abstract]:Objective: to study the acceptability and stability of midazolam syrup mixture prepared by our hospital in Chinese population. Methods: midazolam hydrochloric acid injection (5mg / ml) was mixed with single syrup (850mg / L) to produce 5 groups of different concentrations of midazolam syrup (control group 5 mg / ml M: M: 1: 0A). The mixture of midazolam syrup was titrated in random order to taste the tongue root of 40 healthy Chinese adult volunteers. The volunteers gargled the mouthwash for 60 seconds immediately after each taste.
