利福喷丁对大鼠免疫功能影响的研究

【摘要】:目的:了解应用利福喷丁胶囊后能否增加或抑制与清除结核杆菌的免疫功能,并对免疫功能改变的可能机制进行初步研究;方法:1、取雄性SD大鼠70只,体重180±30g,随机分为四组,分别为喂养前10只、喂养一个月、三个月,喂养三个月停喂一个月,后三组均20只,分为实验组(10只)和对照组(10只)。2、喂养前10只自由饲养,不做任何干预,,另外三组实验组用利福喷丁胶囊灌胃,对照组用生理盐水灌胃,余喂养条件相同。3、用流式细胞术检测CD_3~+、CD_4~+、CD_8~+及NK(CD-3CD_(161)~+)细胞计数,用碳粒廓清法检测巨噬细胞的吞噬功能。结果:1.实验组与对照组各指标比较中,喂养一月及喂养三月停喂一月时各指标差异均无统计学意义,喂养3月实验组与对照组各指标比较中,两组间的CD_3~+、CD_8~+、廓清指数、平均吞噬率四指标之间的差异有统计学意义;两组间的CD_4~+、CD_(161)~+、CD_4~+/CD_8~+三指标差异无统计学意义.2.对照组各时间点与空白对照组比较中,两组间CD-3~+、平均吞噬率、廓清指数之间的差异无统计学意义,CD_4~+、CD_8~+、CD_(161)~+、CD_4~+/CD_8~+在喂养一月、喂养三月,喂养三月停喂一月及空白组之间差异均有统计学意义;3.实验组各时间点与空白对照组比较:CD_3~+、CD_8~+、CD_4~+/CD_8~+这三个指标在喂养一月、喂养三月,喂养三月停喂一月及空白组之间差异均有统计学意义,CD_4~+、CD_(161)~+、平均吞噬率、廓清指数这4个指标在各时间点间差异无统计学意义;结论:1.本实验中,实验组与对照组各指标比较中,喂养一月及喂养三月停喂一月时各指标差异均无统计学意义,表明喂养一月时利福喷丁胶囊的药效未达到影响免疫功能的强度,喂养三月停喂一月,可能由于停药后药物在体内的量减少或停药后原有药物随体内代谢清除,对各指标的差异无显著性的改变;2.喂养3月实验组与对照组比较中,CD_3~+、CD_8~+、廓清指数、平均吞噬率4指标差异有统计学意义,实验组CD_3~+、CD_8~+、廓清指数、平均吞噬率的均数±标准差均比对照组增高,且各指标差异有统计学意义,表明随着喂药时间的延长,药物对个别免疫相关指标有一定影响。3.对照组各时间点与空白对照组比较中,除CD_3~+、平均吞噬率、廓清指数外,指标CD_4~+、CD_4~+/CD_8~+、CD_(161)~+、在喂养一月、喂养三月、喂养三月停喂一月及空白组之间差异均有统计学意义,该差异可能为样本含量小导致的抽样误差或实验操作误差所致;4.实验组各时间点与空白对照组比较中:CD_3~+、CD_8~+、CD_4~+/CD_8~+这三个指标在喂养一月、喂养三月、喂养三月及空白组之间差异均有统计学意义, 表明该药对这些指标的影响与药物作用的时间呈正相关。CD_4~+、CD_(161)~+、平均吞噬率、廓清指数这4指标在各时间点间差异无统计学意义,尚不能排除该药对这些指标无影响。
[Abstract]:Objective: to investigate whether the immune function of Mycobacterium tuberculosis can be increased, inhibited and eliminated by rifapentin capsule, and to study the possible mechanism of immune function change. Methods: 70 male SD rats, weighing 180 卤30 g, were randomly divided into four groups. Ten rats were fed before, one month, three months, one month after feeding, 20 rats in the last three groups were divided into experimental group (n = 10) and control group (n = 10). 10 rats were fed freely without any intervention before feeding. The other three groups were fed with Rifopentin capsule, while the control group were fed with normal saline under the same feeding conditions. Flow cytometry was used to detect the count of CD3 ~ + CD4 ~ + CD4 ~ + and NKT CD-3CDD ~ (161 ~ +) cells, and the phagocytic function of macrophages was detected by carbon particle clearance method. The result is 1: 1. There was no significant difference in each index between the experimental group and the control group after feeding for one month and after feeding for three months. In the comparison of the indexes of the experimental group and the control group, the CD3 ~ + CD8 ~, and the clearance index between the two groups were not statistically significant. There was significant difference among the four indexes of average phagocytosis rate, and there was no significant difference in CD4 / CD4 / CD8 ~ 3 between the two groups. There was no significant difference in CD-3 ~, average phagocytosis rate and clearance index between the two groups at each time point compared with the blank control group. There was no statistical significance between the two groups. There were significant differences between the three months of feeding and the blank group. Compared with the blank control group, there were significant differences in the three indexes of CD4 ~ / CD8 ~ between the control group and the control group. The average phagocytosis rate was significantly higher than that in the control group after feeding for one month, three months after feeding for one month, and between the blank group and the control group, and the mean phagocytosis rate was higher than that in the control group, and the mean phagocytic rate was higher than that in the control group. There was no significant difference between the four indexes at different time points. Conclusion: 1. In this experiment, there was no significant difference in the indexes between the experimental group and the control group after feeding for one month and at the end of feeding for three months, which indicated that the efficacy of Rifopentin capsule did not reach the intensity of influence on immune function after feeding for one month. After feeding for three months or one month, there may be no significant difference between the two indexes because of the decrease in the amount of drug in the body after the withdrawal or the elimination of the original drug with the metabolism of the body after the withdrawal of the drug. Three months after feeding, there were significant differences in CD3 ~ + CD8 ~, clearance index and average phagocytosis rate between the experimental group and the control group. In the experimental group, CD3 ~ + CD8 ~, clearance index and mean 卤standard deviation of the average phagocytosis rate were higher than those in the control group. The difference of each index was statistically significant, which indicated that with the prolongation of the feeding time, the drug had a certain influence on some immune-related indexes. In comparison between the control group and the blank control group at each time point, except for CD3, average phagocytosis rate and clearance index, the index CD4 ~ / CD4 ~ / CD8 ~ + CDT was significantly different between the control group and the control group. There were significant differences between the control group and the control group in feeding for one month, feeding for three months, feeding for one month, and the blank group. The difference may be caused by sampling error caused by small sample size or experimental operation error. Compared with the blank control group at each time point in the experimental group, there were statistically significant differences among the three indexes of the ratio of CD4 / CD8 to CD4 / CD8 ~ in the control group and the control group for the first month, the third month of feeding and the blank group. The results showed that the influence of the drug on these indexes was positively correlated with the time of the drug action. The average phagocytosis rate and clearance index of the four indexes were not statistically significant, and it could not be ruled out that the drug had no effect on these indexes.
