纳米材料功能配基的制备及其应用

【摘要】:目前,纳米材料已逐渐成为了多门学科研究的基础。纳米材料由于其自身的表面效应、小尺寸效应和宏观量子隧道效应等,使其在光学、电学、磁学等方面得到了极为广泛的应用。近来,半导体聚合物纳米颗粒(SPNs)因为它们作为荧光探针的突出性质如高量子产率、大的光子吸收截面积以及良好的光稳定性,已经引起了人们越来越大的关注。 本文通过合成不同两性聚合物配基,并以此作为保护剂,结合纳米沉降等方法,探索开发出水溶性半导体聚合物荧光纳米探针。综合光谱和显微表征表明,该SPNs具有卓越的光物理学性能,如优异的荧光强度,相比量子点减少的光闪烁,比类似大小的荧光蛋白更好的光稳定性。更重要的是,由于光学性能的提高,通过把膜生物标记物交联到SPNs的表面,发现它们不仅适用于活细胞标记,而且也适用于单粒子追踪。具体研究工作主要包括以下几个方面: 1.优化荧光纳米颗粒的合成路线,实现小尺寸荧光纳米颗粒的合成,首先通过合成不同的两性聚合物配基,然后以此作为保护剂,结合纳米沉降等方法,利用[(9,9-二辛基芴-2,7-二基)-交替-共-(1,4-苯并-{2,1’,3}-噻二唑](PFBT)合成水溶性半导体聚合物荧光纳米探针,通过表征发现,合成的纳米粒子具有粒径小、尺寸均匀、荧光强度高等特性。 2.研究合成的半导体聚合物纳米颗粒的光学性能,荧光稳定性,探讨巯基乙胺对其光学性能影响,并在单颗粒层面对其进一步研究。 3.通过所制备的荧光探针颗粒,在活细胞水平上进行肿瘤细胞靶向标记和单颗粒追踪实验研究。
[Abstract]:At present, nanomaterials have gradually become the basis of multi-disciplinary research. Nanomaterials have been widely used in optics, electricity and magnetism due to their surface effect, small size effect and macroscopic quantum tunneling effect. Recently, semiconductor polymer nanoparticles (SPNs) have been characterized by their outstanding properties as fluorescent probes such as high quantum yields, large photon absorption cross-sections, and good photostability. In this paper, different amphoteric polymer ligands were synthesized and used as protective agents to explore the development of water-soluble semiconductor polymer fluorescent nanoprobes. The results show that the SPNs have excellent photophysical properties, such as excellent fluorescence intensity, better photostability than the light scintillation reduced by quantum dots, compared with fluorescent proteins of similar size. More importantly, due to the improvement of optical properties, it is found that the membrane biomarkers are not only suitable for living cell labeling, but also suitable for single particle tracing by cross-linking the membrane biomarkers to the surface of SPNs. The specific research work mainly includes the following aspects: 1. The synthesis route of fluorescent nanoparticles was optimized to realize the synthesis of small size fluorescent nanoparticles. Firstly, different amphoteric polymer ligands were synthesized, and then used as protective agents, combined with nano-deposition, etc. The water-soluble semiconductor polymer fluorescence nanoprobes were synthesized by the method of [Dichlorooctylfluorene -2-dioctylfluorene -7- dimethylfluorene -alternating -co-carbolone 4- benzo-4- {2an1zol} -thiadiazole] PFBT.The characterization results show that the synthesized nanoparticles have the characteristics of small size, uniform size and high fluorescence intensity. 2. The results show that the synthesized nanoparticles have the properties of small size, uniform size and high fluorescence intensity. The optical properties and fluorescence stability of the synthesized semiconductor polymer nanoparticles were studied. The effects of mercaptoethylamine on their optical properties were investigated. The target labeling and single particle tracing of tumor cells were studied on the living cell level by using fluorescent probe particles.
