异常黑胆质成熟剂延缓衰老作用机制的研究

【摘要】:目的:分别观察异常黑胆质成熟剂(ASMq)对D-半乳糖所致衰老模型大鼠学习记忆能力、血液生化指标、免疫功能,心、脑、肝脏组织形态学和心、脑、肝组织中p16蛋白、p-半乳糖苷酶表达以及脾脏NF-κBp65表达的影响,以此进一步阐明ASMq延缓衰老的部分作用机制。方法:选取3个月龄雄性SD大鼠60只,体质量180~220g,采用D-半乳糖制作亚急性衰老模型,在模型建立后分别用高、中、低剂量(6.0,3.0,1.5g/kg) ASMq对衰老大鼠模型进行干预,并于第67d处死大鼠,测定各组主要血常规、血液生化指标,测定各组大鼠血清IL-2、1L-6、TNF-α水平,观察各组大鼠心脏、肝脏、脑组织形态学变化和p16蛋白和β-半乳糖苷酶表达水平以及用半定量RT-PCR法检各组大鼠脾脏NF-κBp65mRNA表达水平。结果:(1)与衰老模型组大鼠相比,正常对照组抵达终点的游泳时间差异有统计学意义(p0.01);衰老模型组与ASMq各给药组组大鼠抵达终点的游泳时间差异有统计学意义(P0.05-0.01)。(2)衰老模型组与ASMq高、中、低剂量组白细胞(WBC)、红细胞(RBC)、血红蛋白(HGB)、红细胞压积(HCT)、平均红细胞体积(MCV)、血小板数目(PLT)、谷丙转氨酶(ALT)、谷草转氨酶(AST)、总蛋白(TP)、血肌酐(CRE)、血尿酸(UA)、白蛋白(ALB)、血糖(GLU)差异无统计学意义(P0.05),但甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)差异有统计学意义(P0.05)。(3)与正常对照组比较,衰老模型组IL-2、IL-6、TNF-α含量差异有统计学意义(P0.01);与衰老模型组比较,ASMq高剂量组IL-2含量升高,IL-6、TNF-a含量降低,差异有统计学意义(P0.01);ASMq中剂量组IL-2含量升高,IL-6、TNF-α含量降低,差异有统计学意义(P0.05-0.01);ASMq低剂量组IL-2含量升高,IL-6含量降低,差异有统计学意义(P0.05),TNF-α含量降低,但差异无统计学意义(P0.05)。(4)心脏:与衰老模型组相比,ASMq各给药组组心肌细胞饱满,肌丝排列整齐,但也出现了轻度血管扩张;脑:与衰老模型组相比,ASMq高、中剂量组细胞形态结构基本完整,排列整齐,但也有轻度的细胞肿胀,ASMq高、中剂量组细胞形态结构基本完整,排列整齐,但也出现了不同程度的的细胞肿胀。ASMq低剂量组出现了少量的空泡样变化和核固缩;肝脏:与衰老模型组相比,ASMq各给药组肝细胞结构完整,排列整齐。有少量的脂质空泡和水肿,但没有出现核固缩。中、低剂量出现了脂质空泡和水肿,低剂量出现了核固缩(5)各组大鼠心脏、肝脏、脑组织中p16蛋白、β-半乳糖苷酶表达阳性的细胞表现为细胞浆染成黄褐色。与正常对照组比较,衰老模型组心、脑、肝组织中的p16蛋白、β-半乳糖苷酶的表达明显升高,差异有统计学意义(P0.05~0.01);与衰老模型组比较,ASMq高、中、低剂量组心、脑、肝组织中p16蛋白、β-半乳糖苷酶的表达明显下降,差异有统计学意义(P0.05~0.01)。(6)与正常对照组比较,衰老模型组大鼠脾脏NF-KBp65mRNA表达水平明显升高,差异有统计学意义(P0.01);与衰老模型组比较,ASMq高、中剂量组脾脏NF-KBp65mRNA表达水平降低,差异有统计学意义(P0.05-0.01),低剂量组脾脏NF-KBp65mRNA表达水平降低,但差异没有统计学意义(P0.05)。结论:本实验得出异常黑胆质成熟剂具有延缓机体衰老作用,其作用机理可能是降低p16蛋白、β-半乳糖苷酶的表达以及下调NF-KBp65mRNA表达水平有关。
[Abstract]:Objective : To investigate the effects of ASMq on the learning and memory ability , blood biochemical index , immune function , heart , brain , liver tissue morphology and the expression of NF - 魏B P65 in rats with D - galactose induced by D - galactose .
There was no significant difference in swimming time between aging model group and ASMq group ( P0.05 - 0.01 ) . ( 2 ) There was no significant difference between aging model group and ASMq group ( P0.05 ) , but triglyceride ( TG ) , total cholesterol ( TC ) , high density lipoprotein ( HDL ) and low density lipoprotein ( LDL ) were statistically significant ( P0.05 ) .
Compared with the aging model group , the levels of IL - 2 , IL - 6 and TNF - a in ASMq high - dose group were decreased , and the difference was significant ( P0.01 ) .
The levels of IL - 2 , IL - 6 and TNF - 伪 in ASMq decreased significantly ( P0.05 - 0.01 ) .
The levels of IL - 2 and IL - 6 in the low - dose ASMq group were higher than those in the ASMq low - dose group ( P0.05 ) , but there was no statistical significance ( P0.05 ) .
Brain : Compared with the aging model group , ASMq was high , the morphological structure of medium - dose group was basically intact , the arrangement was orderly , but also mild cell swelling , ASMq was high , the morphological structure of medium - dose group was basically intact , the arrangement was orderly , but also the cell swelling of different degree appeared .
