过氧化氢通过内质网应激信号通路对Hela细胞自噬和凋亡的诱导作用

【摘要】:目的:利用过氧化氢(H2O2)构建人宫颈癌Hela细胞体外氧化应激模型,观察其对Hela细胞自噬和凋亡的影响,并探讨H2O2与内质网应激(ERS)信号通路之间的关系。方法:以1mmol·L-1 H2O2构建体外氧化应激模型;实验分为对照组、H2O24h组、H2O28h组和H2O212h组,各组细胞分别以H2O2作用0、4、8及12h后,利用Western blotting法检测各组Hela细胞中ERS及凋亡、自噬相关蛋白表达。结果:与对照组比较,各H2O2组Hela细胞中Cleaved Caspase-3表达水平升高(P0.05),Cleaved Caspase-4表达水平亦明显升高(P0.05);各H2O2组Hela细胞中LC3-Ⅱ表达水平高于对照组(P0.05),GRP78表达水平亦高于对照组(P0.05);随着H2O2作用时间的延长,各H2O2组Hela细胞中IRE1a表达水平明显升高(P0.01),p-JNK表达水平明显高于对照组(P0.05)。结论:在H2O2所构建的体外氧化应激模型中H2O2能够诱导Hela细胞发生自噬和凋亡,且该诱导作用由ERS信号通路介导。
[Abstract]:Aim: to investigate the effects of hydrogen peroxide (H _ 2O _ 2) on autophagy and apoptosis of human cervical cancer Hela cells in vitro, and to explore the relationship between H2O2 and endoplasmic reticulum stress (ERS) signaling pathway. Methods: 1mmol L-1 H 2O 2 was used to construct oxidative stress model in vitro, and the cells were divided into two groups: control group (H 2O 2 24 h), H 2O 2 group (H 2O 2 28 h) and H 2O 2 group (H 2O 2 12 h).
