精制橘皮果胶的结构分析

【摘要】:【目的】分析精制橘皮果胶(purified low molecular citrus pectin,PLMCP)的结构,为靶向性抗癌药物载体的研究提供参考。【方法】以橘皮果胶为原料,制备精制橘皮果胶,采用改良间羟联苯法测定其半乳糖醛酸含量,酸碱滴定法和气相色谱法测定其酯化度,分子排阻色谱法测定其分子质量,单糖乙酰化法测定其单糖含量,分析精制橘皮果胶的主要结构。【结果】精制橘皮果胶半乳糖醛酸含量为95.40%,酯化度为7.99%,分子质量在13.69~24.58ku,重均分子质量为17.94ku;单糖只检出半乳糖醛酸,鼠李糖、阿拉伯糖、木糖、葡萄糖和半乳糖均未检出,推测PLMCP为直链结构同型聚半乳糖醛酸。【结论】精制橘皮果胶的结构单一、分布均一、分子质量稳定,可作为新型靶向性抗癌药物的载体。
[Abstract]:[objective] to analyze the structure of refined orange peel pectin (purified low molecular citrus) and to provide a reference for the study of targeted anticancer drug carrier. [methods] Pectin from orange peel was used as raw material to prepare refined orange peel pectin. Galacturonic acid content was determined by modified m-hydroxybiphenyl method, esterification degree was determined by acid-base titration and gas chromatography, molecular weight was determined by molecular exclusion chromatography, monosaccharide content was determined by monosaccharide acetylation method. The main structure of refined orange peel pectin was analyzed. [results] the content of galacturonic acid in refined orange peel pectin was 95.40, the degree of esterification was 7.99m, the molecular weight was 13.69m 24.58kuand the average molecular weight was 17.94ku.The monosaccharide only detected galacturonic acid, rhamnose, arabinose and xylose. Both glucose and galactose were not detected. It was speculated that PLMCP was a straight chain polygalacturonic acid. [conclusion] the purified orange peel pectin has a single structure, uniform distribution, stable molecular weight, and can be used as a carrier of new targeted anticancer drugs.
