载氯诺昔康壳聚糖关节腔注射用温敏凝胶的制备及体外释药性观察

【摘要】:目的制备关节腔注射用载氯诺昔康(Lnxc)壳聚糖温敏凝胶,观察其理化性质及体外释药性。方法以壳聚糖、β-甘油磷酸钠(β-GP)为材料制备载Lnxc温敏凝胶,以胶凝温度、时间、通针性为评价指标,优化处方工艺;采用椎板法观察凝胶的流变学性质,采用离心试验、温度试验观察凝胶的稳定性,采用动态膜透析法观察载Lnxc温敏凝胶和Lnxc溶液的体外释药特性。结果载Lnxc壳聚糖温敏凝胶的最佳处方为Lnxc 2 mg、3%壳聚糖、60%β-GP、p H 7.2,所制备的温敏凝胶属于非牛顿流体,离心后无分层现象,4℃和25℃条件下呈流动状,37℃发生相变形成凝胶。Lnxc溶液8 h药物累积释放率86.70%,载Lnxc温敏凝胶72 h药物累积释放率86.41%,释药规律符合Higuchi方程。结论成功制备载Lnxc壳聚糖关节腔用温敏凝胶,其具有理想的胶凝温度和较好的缓释效果。
[Abstract]:Objective to prepare lornoxicam (Lnxc) chitosan thermosensitive gel for intraarticular injection and observe its physicochemical properties and drug release in vitro. Methods Lnxc thermosensitive gel was prepared by using chitosan and 尾 -GP as materials, the gelation temperature, time and needle permeability were used as evaluation indexes to optimize the prescription process, the rheological properties of the gel were observed by laminar method, and the centrifugal test was used. The stability of the gel was observed by temperature test, and the in vitro release characteristics of Lnxc thermo-sensitive gel and Lnxc solution were observed by dynamic membrane dialysis. Results the best formulation of Lnxc chitosan temperature-sensitive gel was: Lnxc 2 mg / g 3% chitosan 60% 尾 -GPP H 7.2. The temperature sensitive gel was a non-Newtonian fluid.
