伏立诺他对肺腺癌厄洛替尼耐药细胞株耐药性的影响

【摘要】:目的探讨组蛋白去乙酰化酶抑制剂伏立诺他(SAHA)对肺腺癌厄洛替尼(Erlotinib)耐药细胞株PC-9/ER耐药性的影响。方法 PC-9/ER经过厄洛替尼单药、SAHA单药、厄洛替尼联合SAHA处理48 h后,MTT法检测各种药物对PC-9/ER细胞的增殖抑制率;流式细胞术检测细胞凋亡率。结果 SAHA单药对PC-9/ER具有抑制作用,呈明显的剂量依赖效应关系。无细胞毒剂量的SAHA(1μmol/L)能明显提高PC-9/ER对厄洛替尼的敏感性。结论 SAHA能够部分改善PC-9/ER对厄洛替尼的敏感性。
[Abstract]:Objective to investigate the effect of (SAHA), a histone deacetylase inhibitor, on the drug resistance of erlotinib (Erlotinib) resistant lung adenocarcinoma cell line PC-9/ER. Methods after PC-9/ER was treated with erlotinib and SAHA for 48 h, the proliferation inhibition rate of PC-9/ER cells was detected by flow cytometry, and the apoptosis rate was detected by flow cytometry. Results SAHA could inhibit PC-9/ER in a dose-dependent manner. No cytotoxic dose of SAHA (1 渭 mol/L) significantly increased the sensitivity of PC-9/ER to erlotinib. Conclusion SAHA can partially improve the sensitivity of PC-9/ER to erlotinib.
