抗坏血酸对大鼠成骨细胞膜片BMP-2mRNA表达的影响

【摘要】:目的:研究抗坏血酸对成骨细胞膜片BMP-2 mRNA表达的影响。方法:原代培养并鉴定大鼠成骨细胞,采用物理刮取法,体外构建成骨细胞膜片;成骨细胞接种后,分别加入15、50、85 mg/L抗坏血酸,连续培养1~2周,RT-q PCR检测成骨细胞膜片BMP-2 mRNA的表达。结果:获得的细胞符合成骨细胞形态学特征特性。可构建管状形态的成骨细胞膜片。组间比较,第1周和第2周成骨细胞膜片中BMP-2 mRNA表达量实验组均高于对照组,以50 mg/L组的表达量最高(第1周P0.05;第2周P0.05);组内比较,各组表达量第2周均高于第1周(P0.05)。结论:抗坏血酸可促进成骨细胞膜片BMP-2mRNA表达。
[Abstract]:Objective: to study the effect of ascorbic acid on the expression of BMP-2 mRNA in osteoblasts. Methods: primary culture and identification of rat osteoblasts were used to construct osteoblast membrane by physical curettage in vitro. After inoculation of osteoblasts, ascorbic acid (ascorbic acid) was added to the osteoblasts for 1 week and 2 weeks, respectively. The expression of BMP-2 mRNA in osteoblasts was detected by RT-Q PCR. Results: the obtained cells accord with the morphological characteristics of osteoblasts. The tubular osteoblast membrane can be constructed. The expression of BMP-2 mRNA in the first and second week of osteoblast in the experimental group was higher than that in the control group, the expression of BMP-2 mRNA in the 50 mg/L group was the highest (P0.05 in the first week; P0.05 in the second week); in the group, the expression of BMP-2 mRNA in the second week was higher than that in the first week (P0.05). Conclusion: ascorbic acid can promote the expression of BMP-2mRNA in osteoblasts.
