青霉素和红霉素对高脂处理小鼠葡萄糖和脂肪代谢的影响
[Abstract]:Objective to investigate the effects of high fat diet combined with low dose antibiotics on glucose and lipid metabolism in mice. Methods four week-old C57BL/6J male mice were randomly divided into 5 groups, 8 in each group, respectively: high fat control group, (Con), penicillin low concentration group (5 mg/L) (PL), penicillin high concentration group (50 mg/L) (PH), erythromycin low concentration group) (5 mg/L) (EL), erythromycin high concentration group (50 mg/L) (EH). High fat diet combined with drugs was used for 90 days and glucose tolerance test was carried out at 75 days. The pathological changes of liver tissue were observed by HE staining and oil red O staining. Serum free fatty acid (FFA), glucose (Glucose), triglyceride (TG), total cholesterol (TC), low density lipoprotein cholesterol (LDL-C) high density lipoprotein cholesterol (HDLC), and liver TG were determined. The changes of gene expression of PPARa and PPARg in liver tissue were analyzed by real-time fluorescence quantitative PCR. Results the histopathological changes and lipid accumulation in liver cells were significantly increased in the antibiotic drug treated group, especially in the EH treated group. Compared with the hyperlipidemia control group, the glucose tolerance was abnormal in the antibiotic drug treatment group, and the serum FFAG glucose TG increased (P0.05) HDL-C showed a downward trend, but no significant difference. The content of TG and the expression of Pparg in liver of high concentration antibiotic treatment group were significantly higher than that of control group (P0.05). Conclusion low dose exposure of penicillin and erythromycin leads to disorder of lipid and glucose metabolism in liver of mice.
