丝素—氧化石墨烯复合膜的细胞毒性研究

【摘要】:目的: 制备丝素-氧化石墨烯复合膜(SF-GO),并评价丝素-氧化石墨烯复合膜的细胞毒性,为后期在该膜上载药及动物实验提供理论依据。 方法: 1.在普通培养瓶中培养小鼠成纤维细胞L-929; 2.制备丝素-氧化石墨烯复合膜; 3.在丝素-氧化石墨烯复合膜上培养L-929细胞; 4.采用位相差显微镜和电子显微镜观察确认L-929细胞的形态及其生长情况; 5.采用MTT比色法检测含有不同浓度氧化石墨烯(0.1wt%.0.2wt%、0.5wt%.1.0wt%)的丝素-氧化石墨烯复合膜在不同时期(1d、3d、5d、7d)对小鼠成纤维细胞L929相对增殖率(RGR)的影响,评价其细胞毒性。 结果: 随着氧化石墨烯浓度的增加,细胞毒性在不断增加(P0.01);随着测定天数的增加,氧化石墨烯的细胞毒性减小虽无统计学意义,但各组总体趋势都在减小;通过透射电镜照片可见各组细胞形态基本正常,细胞生长良好,为长梭形或卵圆形,并可见圆形分裂细胞,细胞折光性强。 结论: 氧化石墨烯浓度≤1wt%时无明显细胞毒性,可用于后续进一步研究。
[Abstract]:Aim: to prepare fibroin-graphene oxide composite membrane (SF-GO) and evaluate the cytotoxicity of fibroin-graphene oxide composite membrane. Methods: 1. Mouse fibroblasts were cultured in ordinary culture flask. Silk fibroin-graphene oxide composite membrane was prepared. L-929 cells were cultured on fibroin-graphene oxide composite membrane. 4. The morphology and growth of L-929 cells were confirmed by phase contrast microscope and electron microscope. The effects of fibroin-graphene oxide composite membrane containing different concentrations of graphene oxide (0.1 wt.0.2 wtand 0.5 wt.1.0 wt%) on the relative proliferation rate (RGR) of mouse fibroblasts L929 at different periods (1 d ~ 3 d ~ 5 d ~ 7 d) were detected by MTT colorimetry. The cytotoxicity of the membrane was evaluated. Results: with the concentration of graphene oxide increasing, the cytotoxicity of graphene oxide increased (P0.01), the cytotoxicity of graphene oxide decreased with the increase of determination days, but the general trend of each group was decreased. Transmission electron microscopy (TEM) photos showed that the cells in each group were normal in shape, good in growth, long fusiform or oval in shape, round mitotic cells and strong refraction.
