α-硫辛酸对糖尿病大鼠视网膜VEGF表达的影响及机制

【摘要】:目的研究α-硫辛酸(α-LA)对糖尿病大鼠视网膜病变及血管内皮生长因子(VEGF)表达的影响及机制。方法选择72只健康的雄性Wistar大鼠作为实验材料,将72只大鼠分为4组:空白对照组(A组)12只,糖尿病模型组(B组)24只,α-LA治疗组(C组)24只,葡萄糖干预组(D组)12只,B~D组大鼠按照60mg/kg的剂量腹腔注射链脲佐菌素(STZ),C组用100mg/kg的α-LA灌胃,D组用5.0%的葡萄糖溶液灌胃。比较各组大鼠的体质量、空腹血糖(FPG)、空腹胰岛素(FINS),稳态胰岛素抵抗指数(HOMA-IR)、VEGF表达、超氧化钠歧化酶(SOD)、谷胱甘肽(GSH)和IL-6水平,比较B~D组大鼠的GR分期。结果72h后,B~D组大鼠模型构建成功,A组大鼠FPG为(4.57±0.15)mmol/L,B组为(21.72±4.28)mmol/L,C组为(21.54±4.96)mmol/L,D组为(21.83±4.77)mmol/L,A组与B、C组比较差异有统计学意义(P0.05);A组大鼠体质量(210.5±5.2)g,B组(211.2±5.7)g,C组(209.8±5.8)g,D组(208.7±3.4)g,4组的差异无统计学意义(P0.05)。4组大鼠4、8、12周的体质量、FPG、FINS、HOMA-IR、VEGF表达、SOD、GSH和IL-6水平差异有统计学意义(P0.05)。12周后,B组、C组和D组大鼠的眼底镜检查GR分期差异有统计学差异(P0.05)。结论α-LA的应用可以有效抑制大鼠视网膜VEGF的表达,这一作用可能与改善胰岛素抵抗、抑制慢性炎症和氧化应激多个环节相关。
[Abstract]:Objective to study the effect and mechanism of 伪-lipoic acid (伪-LA) on the expression of vascular endothelial growth factor (VEGF) and retinopathy in diabetic rats. Methods Seventy-two healthy male Wistar rats were divided into 4 groups: control group (group A, n = 12), diabetic model group (group B, n = 24) and 伪 LA treatment group (group C, n = 24). There were 12 rats in glucose intervention group (group D). Rats in group D received intraperitoneal injection of streptozotocin (STZ), C) with 伪-LA of 100mg/kg according to the dose of 60mg/kg. Group D was given 5. 0% glucose solution. The body weight, fasting insulin (FINS), steady-state insulin resistance index (HOMA-IR), VEGF) expression, superoxide dismutase (SOD), glutathione (GSH) and IL-6 levels were compared in each group, and GR stages of BSD group were compared. Results after 72 hours, the FPG of group A was (4.57 卤0.15) mmol/L,B, (21.72 卤4.28) mmol/L,C, (21.54 卤4.96) mmol/L,D was (21.83 卤4.77) mmol/L,A and group C was (21.83 卤4.77) mmol/L,A. There was no significant difference between group B and group B (P 0.05, 210.5 卤5.2g / g, P 0.05, 211.2 卤5.7g / g, P 0.05). There was no significant difference between group A and group B (209.8 卤5.8g / g, P 0.05). After 12 weeks, there were significant differences in body mass, FPG,FINS,HOMA-IR,VEGF expression, SOD,GSH and IL-6 levels in rats (P0.05). After 12 weeks, there were significant differences in GR staging between group B, group C and group D (P0.05). Conclusion 伪-LA can effectively inhibit the expression of VEGF in rat retina, which may be related to improving insulin resistance, inhibiting chronic inflammation and oxidative stress.
